Microcytosis of red cells and mild anemia are common in thalassemia carriers but those phenotypes are not specific. It is really a challenge for clinical interpretation of those variants. Co-segregation with disease in affected family members or specific phenotypes such as the abnormal Hb H are very helpful to assess the pathogenicity of rare variants. c.244delT was only reported in a 19-year-old woman with mild microcytosis and hypochromia. There was no other information about this variant reported before. We first described the case of this variant compounded with SEA deletion who presented with moderate anemia. Co-segregation analysis was confirmed by Sanger sequencing. Our study gave evidence for predicting the pathogenicity of this rare missense variant.

Background: Genetic variation contributes to atrial fibrillation (AF), but its impact may vary with age. The Research Program contains whole-genome sequencing of data from 100 574 adult participants with linked electronic health records.
Methods: We assessed clinical, monogenic, and polygenic associations with AF in a cross-sectional analysis, stratified by age: <45 years (n=22 290), 45 to 60 years (n=26 805), and >60 years (n=51 659).

Objective: To expand the clinical phenotype associated with MYRF mutations in disorders of sex development (DSDs).
Methods: We present a case of a 17-year-old patient with a female phenotype who presented with primary amenorrhea.
Results: The patient's external genitalia was entirely female in appearance, though there was no opening of vagina below the orifice of urethra.

Multisystem proteinopathy 1 (MSP1) is a rare autosomal dominant disorder caused by mutations in the valosin-containing protein (VCP) gene typically presenting with inclusion body myopathy (IBM), Paget's disease of bone (PDB), frontotemporal dementia (FTD), and amyotrophic lateral sclerosis (ALS). Parkinsonism is a rare feature of MSP1, occurring in 3-4% of cases, with limited post-mortem evidence suggesting neuronal synucleinopathy. We report a case of VCP-related parkinsonism providing the first in vivo demonstration of phosphorylated alpha-synuclein deposition in skin biopsy, a highly sensitive and specific in vivo biomarker of synucleinopathy.

Allosteric proteins play a central role in biological processes and systems. Identifying the biological impact of mutations on allosteric proteins and the phenotypes they influence during disease initiation and progression presents a significant challenge. In theory, computational methods have the potential to facilitate the interpretation of genetic variants in allosteric proteins on a large scale.

Purpose: To describe and compare the ophthalmological and extra-ophthalmological features of patients with Stickler syndrome due to pathogenic variants in COL2A1 and COL11A1.
Design: Retrospective cross-sectional study nested in a multicentric cohort study.
Methods: Records of patients with a confirmed molecular diagnosis of Stickler syndrome followed-up in the ophthalmology department at Necker-Enfants Malades and Cochin University hospitals (Paris) between 2016 and 2024 were retrospectively reviewed.
