The class C orphan G-protein-coupled receptor (GPCR) GPR156, which lacks the large extracellular region, plays a pivotal role in auditory function through G. Here, we firstly demonstrate that GPR156 with high constitutive activity is essential for maintaining auditory function, and further reveal the structural basis of the sustained role of GPR156. We present the cryo-EM structures of human apo GPR156 and the GPR156-G complex, unveiling a small extracellular region formed by extracellular loop 2 (ECL2) and the N-terminus. The GPR156 dimer in both apo state and G protein-coupled state adopt a transmembrane (TM)5/6-TM5/6 interface, indicating the high constitutive activity of GPR156 in the apo state. Furthermore, C-terminus in G-bound subunit of GPR156 plays a dual role in promoting G protein binding within G-bound subunit while preventing the G-free subunit from binding to additional G protein. Together, these results explain how GPR156 constitutive activity is maintained through dimerization and provide a mechanistic insight into the sustained role of GPR156 in maintaining auditory function.

Background: Frailty is a dynamic condition that may affect mental health. This study aimed to investigate the associations of frailty and its changes with the risks of depressive symptoms across multiple regions in aging populations.
Methods: Data were drawn from five cohort studies in the United States, England, Europe, China, and Mexico.
Introduction: The ADHEAR is a non-surgical Bone Conduction Device (BCD) that makes use of an adhesive adapter. While clinical trials have demonstrated its efficacy with regards to audiological performance, safety and compliance, data on real-world paediatric cohorts is scarce.
Methods: This retrospective cohort study analysed data from paediatric patients fitted with ADHEAR at a tertiary centre between January 2017 and September 2024.

Background: Multi-canal benign paroxysmal positional vertigo (MC-BPPV) is a rare form of BPPV characterized by the simultaneous involvement of multiple semicircular canals, often resulting in complex diagnostic and therapeutic challenges. While MC-BPPV most commonly affects the posterior and horizontal semicircular canals, tri-canal involvement - especially with bilateral asymmetry - is exceedingly rare.
Purpose: To describe a unique case of tri-canal BPPV involving right posterior and horizontal semicircular cupulolithiasis and left horizontal semicircular canalolithiasis, successfully managed with a structured, sequential protocol of repositioning maneuvers including a novel Bascule maneuver.

Purpose: This study investigated the effects of age-related hearing decline on functional networks using resting-state functional magnetic resonance imaging (rs-fMRI). The main objective of the present study was to examine resting-state functional connectivity (RSFC) and graph theory-based network efficiency metrics in 49 adults categorized by age and hearing thresholds to identify the neural mechanisms of age-related hearing decline.
Method: Forty-nine adults with self-reported normal hearing underwent pure-tone audiometry and rs-fMRI.

Misophonia is a condition characterized by intense negative emotional reactions to trigger sounds and related stimuli. In this study, adult listeners (N = 15) with a self-reported history of misophonia symptoms and a control group without misophonia (N = 15) completed listening judgements of recorded misophonia trigger stimuli using a standard scale. Participants also completed an established questionnaire of misophonia symptoms, the Misophonia Questionnaire (MQ).
