Digital health technology (DHT) describes tools and devices that generate or process health data. The application of DHTs could improve the diagnosis, treatment, and surveillance of bacterial infection and the prevention of antimicrobial resistance (AMR). DHTs to optimise antimicrobial use are rapidly being developed. To support the global adoption of DHTs and the opportunities offered to optimise antimicrobial use consensus is needed on what data are required to support antimicrobial decision making. This Series paper will explore bacterial AMR in humans and the need to optimise antimicrobial use in response to this global threat. It will also describe state-of-the-art DHTs to optimise antimicrobial prescribing in high-income and low-income and middle-income countries, and consider what fundamental data are ideally required for and from such technologies to support optimised antimicrobial use.

Background: Respiratory tract infection tests are increasingly available in primary care and are expected to expand in the future. However, there is limited understanding of clinicians' views on their benefits and how to effectively integrate them into daily clinical practice.
Objectives: The aim of this study was to explore healthcare professionals' views on using respiratory tract infection tests to safely reduce unnecessary antibiotic prescriptions for respiratory tract infections in primary care based on their experience in routine practice.

Salmonella enterica serovar Typhi, the etiological agent of Typhoid fever, remains a critical public health concern associated with high morbidity in many developing countries. The widespread emergence of multidrug-resistant (MDR) Salmonella Typhi strains against the fluoroquinolone group of antibiotics, particularly ciprofloxacin, poses a significant global therapeutic challenge with underlying resistance due to mutations in quinolone-resistance determining region (QRDR) of gyrA gene, encoding DNA gyrase subunit A (GyrA). In pursuit of alternative therapeutic candidates, the present study was designed to evaluate ciprofloxacin analogues against prevalent GyrA mutations (S83F, D87G, and D87N) to overcome fluoroquinolone resistance through machine learning (ML)-based approach.

BACKGROUNDThe COVID-19 pandemic has challenged efforts to optimise rational antibacterial use due to uncertainties in treatment protocols.AIMWe investigated the impact of COVID-19 on hospital antibacterial consumption in Belgium from 2017 to 2022, relative to the general and hospitalised population.METHODSWe analysed national reimbursement data using defined daily doses (DDD) and three metrics: DDD/1,000 inhabitants/day (DID), DDD/1,000 patient days (PD) and DDD/1,000 admissions.

Early appropriate antibiotic treatment is vital in reducing patient mortality. However, current antimicrobial susceptibility testing (AST) requires 16-24 hours of incubation, delaying appropriate antibiotic treatment. Flow cytometry (FCM) is a rapid method in assessing fluorescence (such as fluorophores for ROS) at single-cell resolution.

Background: The introduction of posttransplant cyclophosphamide (PTCy) is one of the major achievements in the field of haploidentical stem cell transplantation (haplo-HCT). The transplant conditioning intensity (TCI) score is a refined classification of conditioning regimens that assigns weight scores to conditioning regimen components. The aim of our analysis was twofold: to assess the effect on transplant outcomes of combining PTCy with calcineurin inhibitor + mycophenolate mofetil (MMF) instead of mTOR inhibitor + MMF for GvHD prophylaxis, and to assess the effect of stratification by conditioning intensity in the setting of haplo-HCT.
