Article Abstract

Chromosome numbers and morphology are important characteristics of a species and its evolution. Root tips are the most commonly used tissue as a source of actively dividing cells for chromosome visualization in plants. Previously, rapidly growing root tips were collected from germinating kernels or from seedlings growing in pots or fields. However, the use of adventitious roots (ARs) derived from aerial tissue as meristems for chromosome visualization has always been overlooked. Here, we successfully induced ARs in 12 materials that were investigated, with the exception of . Using ARs meristem we obtained high-quality chromosome spreads for , , , , . , . × . , , . , , , and . The results reported here demonstrate that layering is an alternative and effective method for producing meristematic cells for high-quality chromosome preparation in plant species producing ARs. For species that produce ARs by layering, this protocol is particularly valuable for the development of cost-effective and high-throughput non-invasive cytogenetic studies.

Rationale: Weaver syndrome is a rare congenital overgrowth disorder characterized by a wide spectrum of clinical manifestations that often overlap with other overgrowth syndromes. It is primarily caused by pathogenic variants in the Enhancer of Zeste Homolog 2 (EZH2) gene on chromosome 7q36.1.

Forensic DNA phenotyping has become an increasingly important technology for narrowing down potential stain donors in forensic investigations. Over the past decade, the number of genetic markers required for reliable predictions has steadily increased, posing significant challenges for molecular genetic assay designs. These parallel designs often consume valuable DNA extracted from forensically relevant materials.

Advances in sequencing techniques have revealed extreme chromosomal diversity and underscored the importance of chromoanagenesis in macroevolution, speciation, and the context of cancer and tumor progression. Evolution is essential for adapting to the environment, preparing for future pressures, ensuring survival, and generating diversity. Here, we discuss some events like chromothripsis and other chromosomal rearrangements, emphasizing their roles as driving forces in genomic evolution from cancer to speciation.

Congenital cataracts (CCs) are a leading cause of preventable childhood blindness, with genetic factors playing a crucial role in their etiology. Nance-Horan syndrome (NHS) is a rare X-linked dominant disorder associated with CCs but is often underdiagnosed due to variable expressivity, particularly in female carriers. : This study aimed to explore the genetic landscape of CCs in a Swiss cohort, focusing on two novel and one novel variants and their phenotypic presentation.

Using an electron microscope, thick (30-100 nm wide), linear (not branched), cross-striated protein fibrils with an axial repeat of about 65 nm were detected in mammalian cell nuclei. These fibrils differ from the thin filaments of the nuclear matrix described in the literature. Therefore, in this work, the main efforts were aimed at demonstrating the nuclear origin of thick fibrils.
