The two major molecular subtypes of pancreatic adenocarcinoma reportedly have differential response to FOLFIRINOX-based therapy. To promote rapid assignment of basal versus classical subtypes, an array-based single-sample classifier assay was developed and applied to 74 formalin-fixed, paraffin-embedded biopsy or resection specimens of known subtype based on transcriptomics. The Purity Independent Subtyping of Tumors (PurIST) algorithm assigns subtype based on relative expression of 16 RNAs counted by RNA sequencing (RNAseq) versus more practical array-based NanoString nCounter Elements XT technology. Subtype calls were largely concordant between RNAseq and array methods (72/74, 97% agreement). Compared with the lengthy RNAseq protocol, the array-based assay takes just 3 working days to analyze, permitting rapid reporting of tumor subtype. In conclusion, the PurIST pancreatic cancer classifier has robust performance to classify pancreatic adenocarcinoma into basal versus classical subtypes. Clinical validation studies are underway to evaluate outcome in patients whose standard-of-care chemotherapy regimen is selected on the basis of rapid subtype assignment (NCT04683315).

Background: Breast cancer is a systemic disease, yet the impact of tumor molecular subtype and disease stage on the systemic immune landscape remains poorly understood. In this study, we comprehensively analyzed the systemic immune landscape in a large cohort of breast cancer patients, encompassing all molecular subtypes and disease stages, alongside a control group of healthy donors.
Materials And Methods: Using multi-parameter flow cytometry, we assessed the abundance, phenotype, and activation status of diverse innate and adaptive immune cell populations across peripheral blood samples from 355 breast cancer patients and 65 healthy donors.

Ribonucleotide reductase (RR) is the rate-limiting enzyme for NTPs conversion into dNTPs, playing a central role in genome replication and maintenance. It is composed by two catalytic (RRM1) and two regulatory (alternatively RRM2 and p53R2) subunits, of which RRM2's functionality depends on a diferric center in the active site and is one of the most expressed genes in many tumors, among which Rhabdomyosarcoma (RMS), a rare and aggressive pediatric tumor. Didox (3,4-dihydroxy-benzohydroxamic acid) is a highly effective RRM2 inhibitor with iron chelating properties which shows fewer in vivo side effects than classical RR inhibitors.

Extranodal natural killer/T-cell lymphoma (ENKTCL), nasal type, is a rare and aggressive subtype of non-Hodgkin's lymphoma that often presents with extranodal involvement. While relapse patterns commonly involve the nasal cavity, lungs or skin, cardiac involvement is rare. We report a late 50-year-old patient with ENKTCL, nasal type, who achieved complete remission following chemotherapy and autologous stem cell transplantation.

The chick embryo ventricular cardiomyocyte model provides students easy access to experiments involving fundamental features of cardiac cell physiology and pharmacology. Using standard physiology teaching laboratories and basic cell culture equipment, spontaneously beating colonies of electrically-connected cardiomyocytes can be obtained by the students themselves. Students learn, aseptic techniques and cell culture alongside experiments illustrating, at the simplest level of experimentation, how beating rate can be altered physiologically or pharmacologically.

Pregnancy in women with Ehlers-Danlos syndrome (EDS) carries elevated risks, including prematurity, hemorrhage, and maternal morbidity, posing significant anesthetic challenges. We present the case of a 36-year-old woman with classical EDS (cEDS) and multiple comorbidities, including postural orthostatic tachycardia syndrome, bronchial asthma, congenital adrenal hypoplasia, and chronic venous thrombosis, who underwent an elective cesarean section. A multidisciplinary team developed a comprehensive perioperative plan featuring ultrasound-guided spinal anesthesia, extended post-anesthesia observation, and coordinated recommendations from cardiology, hematology, endocrinology, pulmonology, and other specialties.
