Mucogingival surgery has been widely used in soft gingival tissue augmentation in which autografts are predominantly employed. However, the autografts face grand challenges, such as scarcity of palatal donor tissue and postoperative discomfort. Therefore, development of alternative soft tissue substitutes has been an imperative need. Here, we engineered an interconnected porous bovine serum albumin methacryloyl (BSAMA: B, as a drug carrier and antioxidant)/gelatin methacryloyl (GelMA: G, as a biocompatible collagen-like component)-based cryogel with L-Arginine (Arg) loaded as an angiogenic molecule, which could serve as a promising gingival tissue biohybrid scaffold. BG@Arg cryogels featured macroporous architecture, biodegradation, sponge-like properties, suturability, and sustained Arg release. Moreover, BG@Arg cryogels promoted vessel formation and collagen deposition which play an important role in tissue regeneration. Most interestingly, BG@Arg cryogels were found to enhance antioxidant effects. Finally, the therapeutic effect of BG@Arg on promoting tissue regeneration was confirmed in rat full-thickness skin and oral gingival defect models. In vivo results revealed that BG@Arg2 could promote better angiogenesis, more collagen production, and better modulation of inflammation, as compared to a commercial collagen membrane. These advantages might render BG@Arg cryogels a promising alternative to commercial collagen membrane products and possibly autografts for soft gingival tissue regeneration.

Angiotensin II (Ang II) releases inflammatory mediators from several cell types. The objective of this study was to investigate the potential of Ang II to induce mRNA expression of inflammatory mediators in primary cultured fibroblast-like cells isolated from gingival and periodontal ligament tissues. A synergistic effect of co-treatment with Ang II and Interleukin-1β (IL1β) on the mRNA expression of inflammatory mediators was explored.

Objectives: This study aimed to compare the efficacy of the full-thickness palatal graft technique (FTPGT) and the coronally advanced flap with subepithelial connective tissue graft (CAF + SCTG) in achieving complete root coverage (CRC) in single gingival recessions (GR).
Methods: Forty healthy patients with a single RT1 GR were randomized into two groups: 20 treated with CAF + SCTG and 20 with FTPGT. Baseline and 12-month measurements of GR, keratinized tissue width (KTW), probing depth (PD), clinical attachment level (CAL), and gingival thickness (GT) were recorded.

Aims: To compare the early wound-healing responses to crosslinked hyaluronic acid enriched with two proline-rich peptides (P2, P6) against unmodified hyaluronic acid and the enamel-matrix derivative (EMD) in a porcine gingival-detachment model.
Methods: In six pigs, defects around premolars were treated with HA, HA + P2, HA + P6 or EMD. After 6 days, the sites were harvested and evaluated using histology, immunohistochemistry, multiplex cytokine assay and untargeted proteomics of the gels, which were examined, informing an integrated multiomics approach analysis.

Periodontitis, a pervasive chronic inflammatory disorder, is distinguished by the progressive degradation of periodontal tissues and alveolar bone. Despite remarkable progress in understanding the pathogenesis of periodontitis, the involvement of TCRαβCD4CD8 T cells, also known as double-negative T (DNT) cells, in the pathophysiology of this disease has not been thoroughly investigated. In this study, we observed a significant reduction in the frequency of TCRαβ DNT cells within the gingival tissues of patients afflicted with periodontitis when compared with healthy individuals.

Orthodontic-induced gingival enlargement (OIGE) affects approximately 15-30% of patients undergoing orthodontic treatment and remains largely unpredictable, often relying on subjective clinical assessments made after irreversible tissue changes have occurred. S100A4 is a well-characterized marker of activated fibroblasts involved in pathological tissue remodeling. This was a cross-sectional precision biomarker study that analyzed gingival tissue samples from three groups: healthy controls (n = 60), orthodontic patients without gingival enlargement (n = 31), and patients with clinically diagnosed OIGE (n = 61).
