The first steps of vision take place in the ciliary outer segment compartment of photoreceptor cells. The protein composition of outer segments is uniquely suited to perform this function. The most abundant among these proteins is the visual pigment, rhodopsin, whose outer segment trafficking involves intraflagellar transport (IFT). Here, we report three major findings from the analysis of mice in which ciliary transport was acutely impaired by conditional knockouts of IFT-B subunits. First, we demonstrate the existence of a sorting mechanism whereby mislocalized rhodopsin is recruited to and concentrated in extracellular vesicles prior to their release, presumably to protect the cell from adverse effects of protein mislocalization. Second, reducing rhodopsin expression significantly delays photoreceptor degeneration caused by IFT disruption, suggesting that controlling rhodopsin levels may be an effective therapy for some cases of retinal degenerative disease. Last, the loss of IFT-B subunits does not recapitulate a phenotype observed in mutants of the BBSome (another ciliary transport protein complex relying on IFT) in which non-ciliary proteins accumulate in the outer segment. Whereas it is widely thought that the role of the BBSome is to primarily participate in ciliary transport, our data suggest that the BBSome has another major function independent of IFT and possibly related to maintaining the diffusion barrier of the ciliary transition zone.

In the eye, the retinal pigment epithelium (RPE) maintains the functionality and welfare of retinal photoreceptors and forms a tight, interlocked structure with photoreceptor outer segments (POSs). The RPE-retina interaction is difficult to recapitulate in vitro, limiting the studies addressing the retinal maintenance functions of the RPE. To overcome this challenge, we constructed a retina-mimicking structure using a soft polyacrylamide hydrogel coated with Matrigel.

Background: Irreversible electroporation (IRE) is a novel anticancer ablative treatment, which has been proposed to enhance the efficacy of chemotherapy in the periphery of the ablated area by capturing chemotherapy intracellularly. The aim of the current trial was to characterize the ablated lesion to get spatial information about the distribution of captured chemotherapy and in extension to assess the a priori probability of efficacy for the combined intervention.
Methods: IRE ablations were performed in five pigs with or without concurrent intravenous bleomycin and gadolinium contrast injection.

Two new copepod species of the species-rich genus , collected from temporary water habitats in Southeast Asia, are described. A critical morphological comparison has revealed that the two new species, from Thailand and from Cambodia, closely resemble their respective congeners, (Brehm, 1923) and (Richard, 1894). However, can be distinguished from other congeners, including , by the following characters of the male P5: (1) the inner margin of the right basis has one small semicircular knob and one triangular knob; (2) the right second exopodal segment has a long, slender accessory spine inserted at the proximal third of the outer margin; and (3) the inner margin of the left exopod has a bilobed saw with a series of large denticles and smaller denticles near the distal end.
