Article Abstract

A HPLC method was developed and validated for the novel combination of rutin (RN) and donepezil (DNP). RN and DNP were simultaneously eluted through a C18 column (Ø 150 × 4.6 mm) with a 60:40 v/v ratio of 0.1% formic acid aqueous solution to methanol at 0.5 ml/min. The purposed method was found linear, selective, reproducible, accurate and precise with percent RSD less than 2. The limit of quantification for RN and DNP was found 3.66 and 3.25 μg/ml, respectively. Validated as per the ICH guidelines, the developed method efficiently quantified RN and DNP co-loaded in DQAsomes (121 nm) estimating matrix effect, release profile, entrapment efficiency, loading efficiency and plasma kinetics.

Alzheimer's disease (AD) is the most prevalent type of dementia, negatively affecting the overall quality of life. Targeting the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway, a regulator of protective genes, offers potential AD therapies. Inhibiting Kelch-like ECH-associated protein 1 (Keap1), which suppresses Nrf2, may help reduce neuronal damage.

Article Synopsis
  • - Alzheimer's disease causes severe memory loss and is a leading cause of disability in older adults, with multiple complex factors contributing to its progression, including the buildup of β-amyloid plaques and neuroinflammation.
  • - Traditional treatments like donepezil only manage symptoms without stopping progression and can have significant side effects, leading researchers to explore natural alternatives for a more holistic approach.
  • - This study found various metabolites in Vitex pubescens bark, with a focus on phenolic compounds, and evaluated its antioxidant properties and effects on acetylcholinesterase, indicating potential therapeutic benefits for Alzheimer’s treatment.

Alzheimer's disease, an intricate neurological disorder, is impacting an ever-increasing number of individuals globally, particularly among the aging population. For several decades phytochemicals were used as Ayurveda to treat both communicable and non-communicable diseases. Acetylcholinesterase (AChE) is a widely chosen therapeutic target for the development of early prevention and effective management of neurodegenerative diseases.

Background: Tetrapleura tetraptera (TT) and Quassia undulata (QU) are two predominant tropical ethnobotanicals with various medicinal values but are commonly used in folklore for the treatment of mental illness without justifiable mechanisms of action.

Aim Of The Study: To investigate the effects of aqueous extracts from TT fruits and QU leaves on the spatial and non-spatial working memory, antioxidant status and activities of neuronal marker enzymes of scopolamine-induced amnesic rats and thus, understand the possible mechanism of action of these plants.

Materials And Methods: Fifty-five albino rats were divided into eleven groups.
