Soluble MFGE8 mediates cell entry of Crimean-Congo hemorrhagic fever virus.

mBio

Unlabelled: Crimean Congo hemorrhagic fever virus (CCHFV) causes fatal tick-borne disease in humans and is a priority pathogen of the World Health Organization. No licensed vaccines or specific antiviral drugs are available. To understand the cell entry of CCHFV and identify potential antiviral targets to combat the disease, here, we perform the CRISPR knockout screen in wild-type cells, followed by a complementary CRISPR activation screen in cells deficient in common attachment factors (heparan sulfate, AXL, TIM-1).

lncRNA-gm33782 exacerbates renal inflammation and injury in AKI by activating M1 macrophage polarization through Ccl5/NF-κB signaling.

Acute kidney injury (AKI) progression is closely linked to dysregulated macrophage activation, but the role of tubular-derived lncRNAs in modulating macrophage-driven inflammation remains poorly understood. Here, we identified ‌lncRNA-gm33782‌ as a potentially pathogenic mediator in AKI due to its strongly upregulated expression in ischemia-reperfusion injury (IRI) and cisplatin (Cis) induced AKI mouse models. Interestingly, in Cis-AKI model, renal-specific knockdown of ‌lncRNA-gm33782 via in vivo electroporation significantly attenuated tubular injury and macrophage infiltration.

Zika virus (ZIKV) infection can lead to a variety of clinical outcomes, including severe congenital abnormalities. The phosphatidylserine receptors AXL and TIM-1 are recognized as critical entry factors for ZIKV in vitro. However, it remains unclear whether and how ZIKV regulates these receptors during infection.

This study explored the protective effects of hesperidin (HES) against ifosfamide (IFA)-induced nephrotoxicity in rats using biochemical, histopathological, and immunohistochemical techniques. The rats were administered IFA at a dose of 500 mg/kg body weight, followed by oral HES treatment at doses of 200 and 300 mg/kg body weight. The results demonstrated that HES significantly reduced IFA-induced increases in urea and creatinine levels, indicating improved kidney function.

Chikungunya virus (CHIKV) is an arbovirus that can lead to chronic arthritis and significantly diminish the quality of life of patients. Given the expanding global prevalence of CHIKV and the absence of specific antiviral therapies, there is an urgent need to explore effective treatment options. This study aimed to evaluate the antiviral effects of shikimic acid (SA) against CHIKV through a combination of network pharmacology, molecular docking, and assays.
