N-glycosylation is essential for protein folding in the endoplasmic reticulum (ER). Glycan attachment facilitates the binding of newly synthesised polypeptides to calnexin and calreticulin, two ER-resident lectins that act as chaperones and promote folding. The regulatory mechanism underlying this process is dictated by the glycan composition, and this study has elucidated the function of mannose trimming in the release of misfolded glycoprotein from ER quality control and subsequent transfer to ER-associated degradation (ERAD) in plants.

Interleukin (IL)-15 is primarily known as a pro-inflammatory and anti-apoptotic cytokine, which stimulates the proliferation and survival of key immunocytes, including macrophages (MACs). Yet, it remains unclear how IL-15 specifically impacts MACs in intact human skin, particularly immunoinhibitory, IL-10-producing/secreting M2 MACs (CD206IL-10). In the current pilot study, we explored this in organ-cultured healthy human eyelid skin in the presence of IFNγ (100 IU/mL) to mimic a pro-inflammatory signaling milieu found in several chronic immunodermatoses.

Ricin, a ribosome-inactivating lectin from seeds, has been used as a bioterrorism agent in multiple cases. While passive immunotherapy with anti-ricin antibodies shows promise in preclinical studies, no approved countermeasure exists. Developing effective monoclonal antibodies (mAbs) is challenging, requiring epitope targeting that ensures neutralization of the two most dominant natural ricin isoforms (D and E).

Objectives: The critical need for minimally invasive biomarkers to stratify non-muscle-invasive (NMIBC) and muscle-invasive bladder cancer (MIBC) prompted our investigation into tumor-specific glycan signatures. We hypothesized that lectin-based glycoprofiling could stratify MIBC and NMIBC via stage-specific glycan remodeling. Leveraging the emerging role of lectin microarrays in glycoprofiling, the first comprehensive plasma-based analysis comparing NMIBC and MIBC glycosylation patterns using a high-density 95-lectin platform was conducted.
