Article Abstract

The self-assembly of peptides and peptide analogues may be exploited to develop platforms for different biomedical applications, among which CEST-MRI (chemical exchange saturation transfer magnetic resonance imaging) represents one of the most attractive techniques to be explored as a novel metal-free contrast approach in imaging acquisitions. A lysine-containing peptide sequence (LIVAGK-NH, named K2) was thus modified by insertion, at the N-terminus, of a peptide nucleic acid (PNA) base, leading to a primary amine suitable for the signal generation. a-K2, c-K2, g-K2 and t-K2 peptides were synthesized and characterized. The c-K2 sequence displayed gelling properties and the Watson and Crick pairing, arising from its combination with g-K2, allowed a significant increase in the mechanical responsivity of the hydrogel. These matrices were able to generate a CEST signal around 2.5 ppm from water and, after assessing their cytocompatibility on GL261 (murine glioma), TS/a (murine breast carcinoma), and 3T3-NIH (murine fibroblasts) cell lines, their capability to work as implants for detection, was proved by intratumor injection in Balb/c mice inoculated with TS/a murine breast cancer cells.

Tea saponin (TS), a pentacyclic triterpenoid extracted from tea trees, is a primary active ingredient of tea. They typically resist bacteria and promote acute wound healing. However, maintaining the long-lasting antibacterial effects of TS during acute wound healing is challenging.

Background: Jing An decoction (JA), a traditional Chinese medicine formulation, has demonstrated notable clinical efficacy in the treatment of Tourette syndrome (TS) over the past two decades. The gut-brain axis (GBA) critically regulates neuroinflammation and neurodevelopmental processes, suggesting its potential as a therapeutic target in TS. However, the mechanisms by which JA alleviates TS via the GBA remain unclear.

Sexual dimorphism-driven differences are overcome in a preclinical vaccine model against .

Currently, no vaccine is available to prevent Chagas disease. Experimental vaccines against () have shown high protection, but their development for humans still requires further study. Additionally, the sexual dimorphism observed in Chagas disease, with greater resistance in women, highlights the need to include both sexes in vaccine research to avoid biases.

PT-112 is a novel small molecule exhibiting promising clinical activity in patients with solid tumors. PT-112 kills malignant cells by inhibiting ribosome biogenesis while promoting the emission of immunostimulatory signals. Accordingly, PT-112 is an authentic immunogenic cell death (ICD) inducer and synergizes with immune checkpoint inhibitors in preclinical models of mammary and colorectal carcinoma.

Monitoring Glycolysis by Endogenous P CEST Magnetic Resonance Imaging.

In this study, we present a novel approach to investigate glycolysis by means of the ¹P CEST technique applied to phosphate-containing substrates at their endogenous concentration. The method relies on the assessment of the saturation transfer (ST) observed on the ¹P signals of inorganic phosphate (Pi) or phosphocreatine (PCr) following the selective irradiation of phosphate groups of endogenous molecules exchanging with ATP, Pi, and indirectly with PCr in enzyme-catalyzed reactions. The concentrations of these substrates often fall below the threshold for direct detection.
