Duck plague (DP) is an acute, contagious and fatal disease, caused by duck enteritis virus (DEV), with worldwide distribution causing several outbreaks and posing severe economic losses. The present study was carried out with a goal of development of a live attenuated cell culture based DP vaccine using an Indian strain of DEV and evaluation of its safety, efficacy along with complete genome analysis. The live attenuated DP vaccine (DPvac/IVRI-19) was developed by serial propagation of a virulent isolate of DEV (DEV/India/IVRI-2016) in the chicken embryo fibroblast (CEF) primary cell culture. Adaptation of DEV in CEF cell culture was indicated by more rapid appearance of cytopathic effects (CPE) and gradual increase of virus titre, which reached up to 10 TCID/mL after 41 passages. The safety, immunogenicity and efficacy of the vaccine were determined by immunization trials in ducklings. The DPvac/IVRI-19 was found to be avirulent and completely safe in the ducklings. Further, the vaccine induced both humoral and cell mediated immune responses and afforded 100% protection against the virulent DEV challenge. A comparison of the whole genome of DPvac/IVRI-19 (MZ911871) and DEV/India/IVRI-2016 (MZ824102) revealed significant number of mutations, which might be associated with viral attenuation. Phylogenetic tree of DEV/India/IVRI-2016 revealed its evolutionary relationship with other DEV isolates, but it formed a separate cluster with certain unique mutations. Thus, with the proven safety and 100% efficacy, the DPvac/IVRI-19 is suitable for large scale production with precisely pure form of vaccine and has potential utility at national and global levels.

High-throughput chromosome conformation capture (Hi-C) provides genome-wide insights into chromatin interactions within the three-dimensional structure of the nucleus, making it a powerful tool for studying genome architecture. Here, we provide a modified in situ Hi-C protocol for small cell numbers, utilizing 50-100 embryonic cells at the 8-cell stage to investigate chromatin organization during bovine early embryonic development. This protocol overcomes the challenges of limited sample availability and offers valuable insights into chromatin dynamics during bovine early embryogenesis.

Tracking the translocation of fluorescent-based reporters at the single-cell level in living mouse embryos requires specialized expertise in mouse embryology and deep computational skills. The present study aimed to (1) evaluate the effects of different concentrations of the polysaccharide extract of Cissus sicyoides (PE-Cs) during in vitro culture of preantral follicles included in goat ovarian tissue on (i) follicular morphology and activation, (ii) ovarian stromal density, (iii) follicular and oocyte diameters, (iv) antioxidant enzymes activity (SOD, CAT, and GPx), (v) quantification of MDA, thiol, and nitrite levels; as well as to (2) measure the total antioxidant capacity of the extract. The ovarian cortex fragments were cultured at 39 °C in a humidified atmosphere with 5% CO for 6 d in alpha-modified minimum essential medium (αMEM) supplemented with insulin, transferrin, and selenium; hypoxanthine; glutamine; and bovine serum albumin, which was called αMEM alone or added of PE-Cs at 20, 40, or 80 µg/mL. At the end of the culture period, a reduction in the percentage of normal follicles in all treatments using PE-Cs compared to fresh control and αMEM.
