Experimental discovery of neuropeptides and peptide hormones is a long and tedious task. Mining the genomic and transcriptomic sequence data with robust secretory peptide prediction tools can significantly facilitate subsequent experiments. We describe the application of various in silico neuropeptide discovery methods for the placozoan Trichopax adhaerens as an illustrated example and a powerful experimental paradigm for cellular and evolutionary biology. In total, 33 placozoan (neuro)peptide-like hormone precursors were found using homology-based BLAST search and repeat-based and comparative evolutionary methods. Some of the discovered precursors are homologous to insulins and RFamide precursors from Cnidaria and other animal phyla.

Background: Although lipoprotein(a) [Lp(a)] level elevation is associated with new-onset heart failure (HF), it is unclear if elevated Lp(a) levels predict cardiovascular events in patients with chronic HF. Thus, we examined the association between Lp(a) levels and adverse cardiovascular outcomes in patients with HF.
Methods And Results: A total of 1088 patients with HF undergoing cardiac catheterization at Emory-affiliated hospitals from 2004 to 2022 were divided into low (<30 mg/dL), intermediate (30-49 mg/dL), and high (≥50 mg/dL) Lp(a) groups.

Peptide hormones, a class of pharmacologically active molecules, have a critical role in regulating energy homeostasis. Prohormone convertase 1/3 (also known as PCSK1/3) represents a key enzymatic mechanism in peptide processing, as exemplified with the therapeutic target glucagon-like peptide 1 (GLP-1). However, the full spectrum of peptides generated by PCSK1 and their functional roles remain largely unknown.

Background: How cardiovascular diseases (CVD) predispose to a higher risk of fragility fractures is not well understood. Both contribute to significant components of disease burden and health expenditure. Poor bone quality, central obesity, sarcopenia, falls, and low grip strength are independent risk factors for hip and other fragility fractures and also for CVD and early death.

Background And Aims: Understanding molecular processes of the early phase of atherosclerotic cardiovascular disease conditions is of utmost importance for early prediction and intervention measures.
Methods: We measured 92 cardiovascular-disease-related proteins (Olink, Cardiovascular III) in 2024 elderly participants of the population-based LIFE-Adult study. We analysed the impact of 27 covariables on these proteins including blood counts, cardiovascular risk factors and life-style-related parameters.
