The oxindole scaffold has been the center of several kinase drug discovery programs, some of which have led to approved medicines. A series of two oxindole matched pairs from the literature were identified where TLK2 was potently inhibited as an off-target kinase. The oxindole has long been considered a promiscuous kinase inhibitor template, but across these four specific literature oxindoles TLK2 activity was consistent, while the kinome profile was radically different ranging from narrow to broad spectrum kinome coverage. We synthesized a large series of analogues, utilizing quantitative structure-activity relationship (QSAR) analysis, water mapping of the kinase ATP binding sites, kinome profiling, and small-molecule x-ray structural analysis to optimize TLK2 inhibition and kinome selectivity. This resulted in the identification of several narrow spectrum, sub-family selective, chemical tool compounds including 128 (UNC-CA2-103) that could enable elucidation of TLK2 biology.

The calibration of the JET x-ray spectrometer is presented. The absolute throughput, diffractor focusing, and instrument function of the spectrometer are presented, and the quality of the ion temperature measurement is re-assessed, particularly at the lower end. The addition of a second diffractor enables the simultaneous measurements of the spectra from H- and He-like nickel, which widens the spatial coverage of the core-ion temperature measurements for high-performance plasmas at a fixed Bragg angle range.

Purpose: This study evaluates 30-day community-acquired pneumonia (CAP) readmission rates dependent on discharge antibiotic selection.
Patients And Methods: This is a retrospective, single-center, observational study of patients discharged with a diagnosis of CAP from July 1st, 2022 through June 30th, 2023. Patients included those empirically treated with ceftriaxone plus azithromycin and with documentation of discharge antibiotics.

It is widely acknowledged that child mortality rates have been higher in rural than urban areas in sub-Saharan Africa (SSA); a phenomenon appreciated as the urban advantage. However, since at least the 1980s, this urban advantage has been narrowing, and in some cases reversing across SSA. While existing studies have primarily focused on establishing this relationship, few clearly define what constitutes urban or rural, with authors using different operationalizations.

This study employs a suite of quantum chemical methods to systematically investigate the photoisomerization mechanism and antioxidant activity of resveratrol (Res) and two key derivatives, Azo-Resveratrol (AzoRes) and Dihydro-Resveratrol (dhRes), thereby elucidating the impact of molecular scaffold modification on their structure-activity relationships. Employing density functional theory (DFT), time-dependent DFT (TD-DFT), spin-flip TD-DFT and multistate complete active space second-order perturbation theory (MS-CASPT2), we investigated the geometric configurations, absorption spectra, photoisomerization pathways, and key antioxidant parameters for all three molecules. The results reveal that the substitution of the CC bond with an NN linkage (AzoRes) induces a bathochromic shift in the absorption spectrum, introduces a low-energy n → π* transition, and facilitates a barrierless photoisomerization pathway.
