Criollo cattle, the descendants of animals brought by Iberian colonists to the Americas, have been the subject of natural and human-mediated selection in novel tropical agroecological zones for centuries. Consequently, these breeds have evolved distinct characteristics such as resistance to diseases and exceptional heat tolerance. In addition to European taurine () ancestry, it has been proposed that gene flow from African taurine and Asian indicine () cattle has shaped the ancestry of Criollo cattle. In this study, we analysed Criollo breeds from Colombia and Venezuela using whole-genome sequencing (WGS) and single-nucleotide polymorphism (SNP) array data to examine population structure and admixture at high resolution. Analysis of genetic structure and ancestry components provided evidence for African taurine and Asian indicine admixture in Criollo cattle. In addition, using WGS data, we detected selection signatures associated with a myriad of adaptive traits, revealing genes linked to thermotolerance, reproduction, fertility, immunity and distinct coat and skin coloration traits. This study underscores the remarkable adaptability of Criollo cattle and highlights the genetic richness and potential of these breeds in the face of climate change, habitat flux and disease challenges. Further research is warranted to leverage these findings for more effective and sustainable cattle breeding programmes.

The Bovine Leukocyte Antigen (BoLA) is one of the most important immunologic and genetic components of susceptibility or resistance to disease for cattle. The BoLA-DR region contains the monomorphic locus BoLA-DRA and three BoLA-DRB loci, with the BoLA-DRB3 gene being the most expressed and polymorphic. Its high variability benefits immunological responses against several diseases.

Rarámuri Criollo (RC) cattle have been raised by the isolated Tarahumara communities of Chihuahua, Mexico, for nearly 500 years, mostly under natural selection and minimal management. RC cattle were introduced to the United States Department of Agriculture-Agricultural Research Service Jornada Experimental Range (RCJER) in 2005 to begin evaluations of beef production performance and their adaptation to the harsh ecological and climatic conditions of the Northern Chihuahuan Desert. While this research unveiled crucial information on their phenotypic plasticity and adaptation, the genetic diversity and structure of the RCJER population remains poorly understood.

Understanding the relationship between Raramuri Criollo cattle (RC) and their microbial ruminal ecosystem will help identify advantageous characteristics of adapted cattle as alternatives to achieve sustainable beef production systems. Our objective was to characterize the rumen microbiome of RC in comparison to Angus and Hereford breeds (European, E) and the cross between them (E × RC). Ruminal fluid was collected from 63 cows in their second productive cycle after grazing in the same paddock for 45 d, in the dry (n = 28) and rain (n = 35) seasons.
