Accurate characterization of the microscopic point spread function (PSF) is crucial for achieving high-performance localization microscopy (LM). Traditionally, LM assumes a spatially invariant PSF to simplify the modeling of the imaging system. However, for large fields of view (FOV) imaging, it becomes important to account for the spatially variant nature of the PSF. Here, we propose an accurate and fast principal components analysis-based field-dependent 3D PSF generator (PPG3D) and localizer for LM. Through simulations and experimental three-dimensional (3D) single-molecule localization microscopy (SMLM), we demonstrate the effectiveness of PPG3D, enabling super-resolution imaging of mitochondria and microtubules with high fidelity over a large FOV. A comparison of PPG3D with a shift-variant PSF generator for 3D LM reveals a threefold improvement in accuracy. Moreover, PPG3D is approximately 100 times faster than existing PSF generators, when used in image plane-based interpolation mode. Given its user-friendliness, we believe that PPG3D holds great potential for widespread application in SMLM and other imaging modalities.

This study develops an integrated X-ray absorption spectroscopy (XAS) photoemission electron microscopy (PEEM) platform on beamline BL09U at the Shanghai Synchrotron Radiation Facility (SSRF), enabling nanoscale characterization of complex materials through energy-resolved imaging and local-area XAS. By using the wide range of energy tunability, full access to different polarizations and PEEM's surface sensitivity, we have established a gap-monochromator control system under the EPICS framework to synchronize the elliptically polarized undulator (EPU) gap and monochromator energy dynamically, optimizing photon flux stability for absorption fine structure analysis. Combining X-ray magnetic circular dichroism (XMCD) and X-ray magnetic linear dichroism (XMLD) with PEEM and local-area XAS, this platform achieves concurrent mapping of electronic structures and magnetic domains in ferromagnetic nano-patterns, as demonstrated through our studies of NiFe Permalloy using this system.

Indocyanine green (ICG) is a well-established near-infrared dye which has been used clinically for several decades. Recently, it has been utilised for fluorescence-guided surgery in a range of solid cancer types, including sarcoma, with the aim of reducing the positive margin rate. The increased uptake and retention of ICG within tumours, compared with normal tissue, gives surgeons a visual reference to aid resection when viewed through a near-infrared camera.

Rationale: Autism spectrum disorders (ASD) are a group of neurodevelopmental and multifactorial conditions with cognitive manifestations. The valproic acid (VPA) rat model is a well-validated model that successfully reproduces the behavioral and neuroanatomical alterations of ASD.
