and are two invasive mosquitoes native to East Asia that are quickly establishing in temperate regions of Europe. Both species are vectors of arboviruses, but we currently lack a clear understanding of their evolution. Here, we present new short-read, shallow genome sequencing of and individuals from northern Italy, which we used for downstream phylogenetic and barcode analyses. We explored associated microbial DNA and found high occurrences of bacteria in both samples, but neither nor We then assembled complete mitogenomes and used these data to infer divergence times estimating the split of from in the Oligocene, which was more recent than that previously reported using mitochondrial markers. We recover a younger age for most other nodes within Aedini and other Culicidae. COI barcoding and phylogenetic analyses indicate that , , and the two sampled from Europe should be considered as separate species within a monophyletic species complex. Our studies further clarify the evolution of and and indicate the need to obtain whole-genome data from putative species in order to disentangle their complex patterns of evolution.

Purpose: Fetal cells in maternal blood are a pure source of fetal genomic DNA for noninvasive prenatal testing (NIPT), if successfully isolated. We assessed whether single-cell genome and transcriptome sequencing (G&T-seq) can be applied to efficiently isolate fetal nucleated red blood cells (fNRBCs) suitable for genetic testing.
Methods: Using umbilical cord blood as a model, we isolated 165 single NRBC candidates from four samples and 12 single lymphocytes as controls from one sample.

Determining the Homologous Recombination Deficiency (HRD)-status of a malignant tumor is central in predicting patient response to specific treatments. Therefore, precise and cost-effective tools are needed for clinical implementation. HRDetect is widely regarded as a golden standard for determining HRD-status.

Background And Aims: There is an urgent need for effective prognostic biomarkers to better stratify patients with advanced hepatocellular carcinoma (HCC) undergoing systemic therapy. Shallow whole-genome sequencing (sWGS) of cell-free DNA (cfDNA) is a cost-effective approach for assessing circulating tumour DNA (ctDNA), genomic alterations, and fragmentomic patterns. This study aimed to evaluate sWGS-derived biomarkers as predictors of outcomes in advanced HCC patients receiving systemic treatment.

Barrett's esophagus (BE), a metaplastic condition that is the only known precursor for esophageal adenocarcinoma (EAC), is relatively common, but progression to cancer is infrequent. BE is inflamed but the contribution of the immune system to the carcinogenic process is unknown. To this end, we contrasted of BE patients, captured when they did not progress (non-progressors), did subsequently, but had not yet progressed (pre-progressors) or had already progressed to EAC (progressors).

Introduction: The molecular and histological characteristics of primary duodenal adenocarcinoma (DA) have been poorly described, which hampers the development of new treatment options. This study aimed to characterize the landscape of chromosomal copy number aberrations (CNAs), microsatellite instability status, and tumor-stroma content, and their association with clinicopathological characteristics of patients with DA.
Methods: DNA was extracted from tumor tissues of patients who underwent a primary surgical resection for DA in a single center (2000-2019).
