Article Abstract

The African naked mole-rat (Heterocephalus glaber) is an attractive model for cancer and aging research due to its peculiar biological traits, such as unusual long life span and resistance to cancer. The establishment of induced pluripotent stem cells (iPSCs) would be a useful tool for in vitro studies but, in this species, the reprogramming of somatic cells is problematic because of their stable epigenome. Therefore, an alternative approach is the derivation of embryonic stem cells from in vitro-produced embryos. In this study, immature oocytes, opportunistically retrieved from sexually inactive females, underwent first in vitro maturation (IVM) and then in vitro fertilization via piezo-intracytoplasmic sperm injection (ICSI). Injected oocytes were then cultivated with two different approaches: (i) in an in vitro culture and (ii) in an isolated mouse oviduct organ culture system. The second approach led to the development of blastocysts, which were fixed and stained for further analysis.

Modulation of prostaglandin synthesis, specifically, decreasing prostaglandin F (PGF) and increasing prostaglandin E (PGE) and interferon-tau (IFNT), can support maternal recognition of pregnancy (MRP) in cattle. Conjugated linoleic acid (CLA) is known to influence prostaglandins (PG) synthesis in cell cultures; however, its effect on in vitro-cultured bovine oocytes and embryos remain unclear. We hypothesized that CLA supplementation in oocytes and embryo culture media would reduce PGF synthesis, increases PGE synthesis and PGE:PGF ratio in embryos, modulate the expression of genes involved in PG and IFNT synthesis, and enhance embryo development.

Fertility traits such as daughter pregnancy rate (DPR), cow conception rate, and heifer conception rate are key predictors of reproductive performance in dairy herds. However, their low heritability, likely due to their multifactorial nature and difficulty in measuring phenotypes, poses challenges for genetic improvement. Oocyte competence, encompassing nuclear and cytoplasmic maturation, is a critical factor influencing fertility.

The aim of this study was to establish a humanized immune system model in severe combined immunodeficient (SCID) mice, assess dendritic cell (DC) phenotype, and evaluate the therapeutic efficacy of a DC-based vaccine in a bladder cancer model. Bladder cancer was induced in SCID mice by injection of T24 cells, followed by human peripheral blood leukocyte (hu-PBL) inoculation to reconstitute the human immune system. DCs were generated in vitro by culturing hu-PBL for 5 days and matured on the eighth day.
