The integrin-associated proteins (IAPs) function in a tightly regulated and coordinated manner to maintain the complex cytoarchitecture at the myotendinous junctions (MTJs) of Drosophila indirect flight muscles (IFMs). Parvin, a conserved but less explored IAP, forms a ternary complex with ILK and PINCH (the IPP complex). Although the IPP complex is functionally conserved, playing a central role in integrin-mediated adhesion, its individual components may also exert independent roles.

Presynaptic active zones (AZs) form during synaptogenesis and are critical for ensuring precise synaptic transmission. Although much is known about the development of AZs at the neuromuscular junction, their assembly in the central nervous system (CNS) remains incompletely understood. Here, we demonstrate the mushroom body α/β Kenyon cells in young adults in which new AZs are continuously formed, as a novel system to dissect the sequential AZ assembly in the CNS.

The Drosophila cell adhesion molecule Sidekick is a key component of tricellular adherens junctions in epithelia, and localizes to specific synaptic layers in the optic lobes. Using mutagenesis of endogenous Sidekick, we showed that its enrichment at apical tricellular junctions and its function in cell rearrangement require its fifth and sixth immunoglobulin domains, but not the first four, although these mediate homophilic adhesion of mammalian Sidekick homologues. The C-terminal PDZ-binding motif of Sidekick contributes to localizing both Sidekick and its intracellular binding partner Canoe to tricellular adherens junctions.

Supracellular actomyosin assemblies: master coordinators of development.

Development

Most movement in biological systems is driven by assemblies of actomyosin, be it in the form of sarcomeres in muscles or as actomyosin networks in non-muscle cells. Actomyosin has several key functions within epithelial cells, the cells that will form most of the organs of an animal during development. One such function is to support cellular shape through an actomyosin cortex just underneath the plasma membrane.

The SAG1-related sequence (SRS) protein family was initially identified in as glycosyl-phosphatidylinositol-anchored surface antigens. More recently, they have been identified in , the causative agent of neosporosis, a leading cause of bovine abortion worldwide. These proteins are implicated in parasite adhesion to and invasion of host cells, immune response modulation and structural roles in the cyst wall.
