Unlabelled: Carrageenan oligosaccharides are important products that have demonstrated numerous bioactivities useful in the food, medicine, and cosmetics industries. However, the specific structure-function relationships of carrageenan oligosaccharides are not clearly described due to the deficiency of high specific carrageenases. Here, a truncated mutant OUC-FaKC16Q based on the reported -neocarratetrose (N4)-producing -carrageenase OUC-FaKC16A from was constructed and further studied. After truncating the C-terminal Por_Secre_tail (PorS) domain (responsible for substrate binding), the catalytic efficiency and temperature stability decreased to a certain extent. Surprisingly, this truncation also enabled OUC-FaKC16Q to hydrolyze N4 into -neocarrabiose (N2). The offset of Arg residue in OUC-FaKC16Q may explain this change. Moreover, the high catalytic abilities, the main products, and the degradation modes of OUC-FaKC16A and OUC-FaKC16Q toward furcellaran were also demonstrated. Data suggested OUC-FaKC16A and OUC-FaKC16Q could hydrolyze furcellaran to produce mainly the desulfated oligosaccharides DA-G-(DA-G4S) and DA-G-DA-G4S, respectively. As a result, the spectrum of products of -carrageenase OUC-FaKC16A has been fully expanded in this study, indicating its promising potential for application in the biomanufacturing of carrageenan oligosaccharides with specific structures.

Introduction: Under Subacute Ruminal Acidosis (SARA) conditions, harmful substances released by the massive lysis of ruminal bacteria are further degraded into small bacterial peptides, such as muramyl dipeptide (MDP). These degradation products are absorbed through the rumen wall and enter the bloodstream continuously, triggering a series of nutritional metabolic diseases and systemic pro-inflammatory responses. Therefore, inhibiting MDP-induced damage emerges as a novel target for preventing and alleviating SARA.

Carrageenan kappa (CK), a polysaccharide obtained from marine red algae, oligo-carragenan kappa (OCK), an oligosaccharide obtained by acid hydrolysis of CK, and oligo-ulvans (OU), an oligosaccharide obtained by acid hydrolysis of ulvans from marine green algae, were sprayed on leaves of Arabidopsis thaliana at a concentration of 1 mg mL, five times, every two days, and plants were cultivated for ten additional days. Plants treated with water (control) or with an aqueous solution of CK, OCK and OU showed an increase in fresh weight (FW) and dry weight (DW), mainly in those treated with OCK and OU. Plants treated with OCK and OU showed an increase in rosette diameter and length of the primary root, but not those treated with CK.

Carrageenans are highly diverse sulfated galactans found in red seaweeds. They play various physiological roles within macroalgae, but also serve as carbon sources for heterotrophic marine bacteria living at their surface. Carrageenan sulfatases catalyze the removal of sulfate esters from the glycans to expose the saccharide chain for further enzymatic processing.

Plant protein-based thermo-reversible gels are essential for meeting the dietary nutritional needs of the elderly and individuals with dysphagia. However, high hydrophobic globulin content and heat-induced denaturation often hinder gel formation.
