Article Abstract

Gene editing has great potential in biomedical research including disease diagnosis and treatment. Clustered regularly interspaced short palindromic repeats (CRISPR) is the most straightforward and cost-effective method. The efficient and precise delivery of CRISPR can impact the specificity and efficacy of gene editing. In recent years, synthetic nanoparticles have been discovered as effective CRISPR/Cas9 delivery vehicles. We categorized synthetic nanoparticles for CRISPR/Cas9 delivery and discribed their advantages and disadvantages. Further, the building blocks of different kinds of nanoparticles and their applications in cells/tissues, cancer and other diseases were described in detail. Finally, the challenges encountered in the clinical application of CRISPR/Cas9 delivery materials were discussed, and potential solutions were provided regarding efficiency and biosafety issues.

The emergence of messenger ribonucleic acid (mRNA) vaccines as an alternative platform to traditional vaccines has been accompanied by advances in nanobiotechnology, which have improved the stability and delivery of these vaccines through novel nanoparticles (NPs). Specifically, the development of NPs for mRNA delivery has facilitated the loading, protection and release of mRNA in the biological microenvironment, leading to the stimulation of mRNA translation for effective intervention strategies. Intriguingly, two mRNA vaccines, BNT162b2 (Pfizer-BioNTech) and mRNA-1273 (Moderna), have been permitted for emergency usage authorization to prevent COVID-19 infection by USFDA.

Polymer particles, including synthetic polymers such as poly(methyl methacrylate) (PMMA) and poly(styrene-co-divinylbenzene) (P(S-co-DVB)) beads, have been widely used as enzymatic supports and drug carriers. In this sense, it is important to understand the stabilization or degradation of such polymer matrices under specific chemical and enzymatic media. Poly(γ-stearyl-l-glutamate) (PSLG) is a semiflexible synthetic polypeptide that forms both thermotropic and lyotropic liquid crystal (LC) phases. We previously showed that spherical nanoparticles (NPs) decorated with another semiflexible helical polymer, poly(hexyl isocyanate), form lyotropic nematic rather than cubic LC phases. A novel electrocatalyst, zirconium ferrite nanoparticles (NPs) (ZrFeO NPs), was synthesized through coprecipitation and calcination processes at 300 °C and 500 °C using iron rust. The ZrFeO NPs were used as catalysts for the hydrogen evolution reaction. Furthermore, these NPs in an alkaline medium exhibited superior properties of a fractional order supercapacitor, based on which a prototype device was fabricated to demonstrate its energy storage applications.
