Nucleoside analogs have served as the cornerstone of antiviral therapy by acting as antimetabolites that disrupt viral DNA or RNA synthesis, thereby effectively inhibiting viral replication. Despite their clinical success, many nucleoside-based antivirals suffer from intrinsic limitations such as poor lipophilicity, low membrane permeability, and rapid metabolic degradation, all of which compromise oral bioavailability and therapeutic efficacy. To address these challenges, lipid conjugation has emerged as a promising prodrug strategy that enhances pharmacokinetic properties, improves cellular uptake, and enables targeted delivery.

LP-98 is a lipopeptide HIV fusion inhibitor showing strong treatment and pre-exposure prophylaxis efficacies in non-human primates. In this study, we further characterized its pharmacokinetics, long-lasting antiviral activity, and post-exposure prophylaxis (PEP) efficacy using 62 macaques. In cynomolgus macaques, LP-98 achieved high concentrations (C) with a half-life (T) of ∼31 h, and sustained an effective therapeutic concentration for two weeks post-injection.

Background And Aims: Sofosbuvir (SOF) plus daclatasvir (DCV) is a primary chronic hepatitis C virus (HCV) treatment in low- and middle-income countries. WHO guidelines recommend a half-adult dose for children (14-25 kg) based on pharmacokinetic modelling, requiring clinical validation. We evaluated the pharmacokinetics, safety, efficacy and acceptability of DCV (30 mg) and SOF (200 mg) in children weighing 14 to < 17 kg and 17-35 kg.

Porcine reproductive and respiratory syndrome virus (PRRSV), an Arteriviridae family enveloped RNA virus, is a major swine pathogen. Using yeast transformation-associated recombination (TAR) cloning, we efficiently generated infectious PRRSV and GFP-expressing clones, identifying transcription-regulating sequences as essential for stable foreign gene expression. Screening SARS-CoV-2 antivirals showed potent inhibition by the multitarget drug ribavirin, the polymerase inhibitors remdesivir and its metabolite GS-441524.
