Article Abstract

Human immunodeficiency virus type 1 (HIV-1), a lentivirus that causes acquired immunodeficiency syndrome (AIDS), poses a serious threat to global public health. Since the advent of the first drug zidovudine, a number of anti-HIV agents acting on different targets have been approved to combat HIV/AIDS. Among the abundant heterocyclic families, quinoline and isoquinoline moieties are recognized as promising scaffolds for HIV inhibition. This review intends to highlight the advances in diverse chemical structures and abundant biological activity of quinolines and isoquinolines as anti-HIV agents acting on different targets, which aims to provide useful references and inspirations to design and develop novel HIV inhibitors for medicinal chemists.

The catalytic asymmetric Mannich reaction is a multicomponent reaction which affords β-amino carbonyl compounds by utilizing an aldehyde, a primary or secondary amine/ammonia, and a ketone. β-amino carbonyl scaffolds are crucial intermediates for the synthesis of naturally occurring bioactive compounds and their derivatives. The synthesized natural compounds exhibit a broad spectrum of biological activities including anti-fungal, anti-cancer, anti-bacterial, anti-HIV, anti-oxidant, and anti-inflammatory activities.

The classical 1,3-dipolar cycloaddition reaction predominantly generates five-membered heterocycles through cycloaddition between 1,3-dipoles and unsaturated hydrocarbons, such as alkenes or alkynes. Herein, we report a light-mediated approach that employs saturated amines as alternatives to conventional unsaturated hydrocarbons, enabling their reaction with -imino(iso)quinolinium ylides to achieve efficient synthesis of pyrazolo-fused (iso)quinoline core structures. This method demonstrates operational simplicity, mild reaction conditions, and excellent functional group compatibility.

The present study is the first report on the development of a set of benzopyridine-fused benzimidazoles, leading to the identification of a lead and its further optimization as a potential preclinical candidate for treating infection. The designed synthetic method for these derivatives is devoid of toxic chemicals and sophisticated reaction setups, using economical and readily available chemicals to produce benzopyridine-fused (namely, quinoline/isoquinoline-fused) benzimidazole derivatives in moderate-to-good yields.

Aromathecin compounds-which contain the same indolizine core structure as camptothecin-like compounds-are expected to show anticancer activity. Among them, 22-hydroxyacuminatine-which has a substituent on the E-ring of the pentacyclic scaffold-exhibits topoisomerase 1 inhibitory activity; therefore, the development of efficient methods for its synthesis has been actively pursued. Herein, we report a versatile synthetic methodology for introducing various substituents on the E-ring, leading to the total synthesis of 22-hydroxyacuminatine as a model compound of the aromathecin family.
