Introduction And Objective: Osmotic changes during the process of freeze-thawing involve changes in the location of aquaporins (AQPs) in membrane domains of spermatozoa. Some AQPs, like aquaporin 3 (AQP3), are linked to sperm cryotolerance in the porcine species. Conspicuous individual variability exists between rams and their ejaculates, which may be classified as displaying good freezability (GFE) or poor freezability (PFE), depending on several endogenous and environmental factors. The present work aimed to examine whether differences in freezability could even involve changes in location and expression of AQP3 in ram spermatozoa.
Methods: Thirty ejaculates from 10 rams (three of each) were evaluated and subsequently classified as GFE ( = 13) or PFE ( = 17) through a principal component analysis (PCA) and k-means cluster analysis. Spermatozoa were examined for the presence, abundance and distribution of AQP3 by western blot and immunocytochemistry, employing a commercial rabbit polyclonal antibody (AQP3 - ab125219).
Results And Discussion: Although AQP3 was found in the sperm acrosome, midpiece, principal and end piece of the tail in both fresh and after frozen-thawed samples, its highest immunolabeling was found in the mid- and principal piece. In the GFE group, the expression of AQP3 in the mid- and principal piece was greater ( < 0.05) in frozen-thawed samples than in fresh specimens while such differences were not detected in the PFE group. Sperm cryotolerance relates to changes in AQP3 expression and thus AQP3 could be used as a biomarker for cryotolerance.
Conclusion: A greater capacity of AQP3 localization in mid- and principal piece of the spermatozoa could be linked to an increase the osmo-adaptative capacity of ejaculates with better capacity to withstand freeze-thawing processes.

Sperm capacitation involves proteolytic remodeling of membrane proteins, including components of the CatSper calcium channel, which is essential for hyperactivation and male fertility. Here, we identify the seminal protease inhibitor SPINK3, a known decapacitation factor that suppresses premature capacitation in the female tract, as the first physiological inhibitor of CATSPER1 processing. In mouse sperm, SPINK3 blocks capacitation-induced CATSPER1 cleavage, preserving a subpopulation with intact CatSper channels and lacking pTyr development in the flagellum.

In the evaluation of male infertility, precise assessment of sperm functional competence has surpassed the requirements of conventional semen parameters. Existing computer-aided analysis systems are deficient at the molecular diagnostic level and also face challenges in live-cell fluorescence quantification. To address these issues, we have developed a novel integrated computational-imaging platform that combines a fine-tuned You Only Look Once version 8 (YOLOv8) architecture, tailored for the EVISEN dataset, with dual-probe fluorescence microscopy image segmentation, enabling simultaneous quantification of intracellular pH (pHi) and mitochondrial DNA G-quadruplexes (mtDNA G4s).

Studies of music-evoked autobiographical memories (MEAMs) show that music is a potent cue for retrieving vivid and self-relevant memories. However, whether and how musical features are able to predict the qualities of MEAMs - including their emotional qualities, phenomenological characteristics and retrieval efficiency - remains unclear. In our study, a sample of 233 adult participants identified a piece of music that evoked an autobiographical memory (AM) before providing a written description of the memory, and then evaluating its emotional and phenomenological content.

The focal "hot spot" neuropathologies in COVID-19 infection are revealing footprints of a hidden underlying collapse of a novel ultrafast ultradian Piezo2 signaling system within the nervous system. Paradoxically, the same initiating pathophysiology may underpin the systemic findings in COVID-19 infection, namely the multiorgan SARS-CoV-2 infection-induced vascular pathologies and brain-body-wide systemic pro-inflammatory signaling, depending on the concentration and exposure to infecting SARS-CoV-2 viruses. This common initiating microdamage is suggested to be the primary damage or the acquired channelopathy of the Piezo2 ion channel, leading to a principal gateway to pathophysiology.

This is also true for Na+,K+-ATPase α1, the other Na+,K+-ATPase isoform present in sperm and also in all cells. However, Na+,K+-ATPase α4 distribution changes when the cells are capacitated, an event necessary for fertilization.
