Glioblastoma (GBM) is the most aggressive brain tumor, with a median survival of ∼15 months. Targeted approaches have not been successful in this tumor type due to the large extent of intratumor heterogeneity. Mosaic amplification of oncogenes suggests that multiple genetically distinct clones are present in each tumor. To uncover the relationships between genetically diverse subpopulations of GBM cells and their native tumor microenvironment, we employ highly multiplexed spatial protein profiling coupled with single-cell spatial mapping of fluorescence in situ hybridization (FISH) for EGFR, CDK4, and PDGFRA. Single-cell FISH analysis of a total of 35,843 single nuclei reveals that tumors in which amplifications of EGFR and CDK4 more frequently co-occur in the same cell exhibit higher infiltration of CD163 immunosuppressive macrophages. Our results suggest that high-throughput assessment of genomic alterations at the single-cell level could provide a measure for predicting the immune state of GBM.

CD151 is a tetraspanin, abnormally expressed in triple negative breast cancer (TNBC). It is a prominent component of exosomes, facilitating the secretion of proteins that promote metastasis and drug resistance. We have previously demonstrated that silencing the CD151 gene reduces metastasis in TNBC.

Background: Although some studies have indicated that CDK4/6 inhibitors are beneficial for the progression-free survival (PFS) and overall survival (OS) in breast cancer, evidence regarding the assessment of clinical response remains insufficient. Therefore, this study aims not only to evaluate the efficacy and safety of CDK4/6 inhibitors combined with endocrine therapy in HR(+)/HER2(-) metastatic breast cancer, but also to analyze the objective response rate (ORR) and clinical benefit rate (CBR), providing comprehensive clinical outcome insights.

Data from cell viability assays, which measure cumulative division and death events in a population and reflect substantial cellular heterogeneity, are widely available. However, interpreting such data with mechanistic computational models is hindered because direct model/data comparison is often muddled. We developed an algorithm that tracks simulated division and death events in mechanistically-detailed single-cell lineages to enable such a model/data comparison and suggest causes of cell-cell drug response variability.

Introduction: Neoadjuvant therapy is the standard of care for the treatment of human epidermal growth factor receptor 2 (HER2)-positive breast cancer (BC). Studies on first-generation antibody-drug conjugates, such as trastuzumab emtansine (T-DM1), showed equal or slightly lower efficacy than chemotherapy combined with dual HER2 blockade. Trastuzumab deruxtecan (T-DXd) is a next-generation conjugate approved for the treatment of metastatic HER2-positive and HER2-low BC, with greatly improved efficacy compared to T-DM1.
