Article Abstract

Right ventricular failure (RVF) is the most important prognostic factor for morbidity and mortality in pulmonary arterial hypertension (PAH) or pulmonary hypertension (PH) caused by left heart diseases. However, right ventricle (RV) remodeling is understudied and not targeted by specific therapies. This can be partly explained by the lack of basic knowledge of RV remodeling. Since the physiology and hemodynamic function of the RV differ from those of the left ventricle (LV), the mechanisms of LV dysfunction cannot be generalized to that of the RV, albeit a knowledge of these being helpful to understanding RV remodeling and dysfunction. Store-operated Ca entry (SOCE) has recently emerged to participate in the LV cardiomyocyte Ca homeostasis and as a critical player in Ca mishandling in a pathological context. In this paper, we highlight the current knowledge on the SOCE contribution to the LV and RV dysfunctions, as SOCE molecules are present in both compartments. he relative lack of studies on RV dysfunction indicates the necessity of further investigations, a significant challenge over the coming years.

Photodynamic therapy (PDT) is a minimally invasive therapeutic modality approved by the Food and Drug Administration (FDA) for the treatment of several pathological conditions, including cancer. Developing new photosensitizers (PSs) for PDT is of great interest to increase the treatment efficacy while minimizing side effects. Rationale: People with cystic fibrosis (pwCF) exhibit chronic and hyperactive neutrophilia which results in a progressive loss of lung function. CF neutrophils have elevated store operated Ca entry (SOCE) relative to healthy non-CF neutrophils, which contributes to persistent neutrophilia. The vast majority of pwCF now take CFTR modulators such as elexacaftor/tezacaftor/ivacaftor (ETI), which effectively increase CFTR function in multiple organs including the lung.

Calcium ions (Ca), pivotal regulators of tumour progression, drive cancer metastasis and therapy resistance via STIM1-mediated store-operated calcium entry (SOCE). This review focuses on the role of STIM1 and SOCE-mediated calcium signalling in various cancer types, with particular emphasis on oral cancer as a key area of our research, and future research directions in these areas are proposed. Specifically, this review reveals the mechanism by which the STIM1/Orai1 complex activates calmodulin (CaM)-dependent effectors to promote epithelial-mesenchymal transition (EMT) and remodelling of the cytoskeleton and immunosuppressive microenvironment.
