Article Abstract

Many bacterial strains are developing mechanism of resistance to antibiotics, rendering last-resort antibiotics inactive. Therefore, new drugs are needed and in particular metal-based compounds represent a valid starting point to explore new antibiotic classes. In this study, we have chosen to investigate gallium(III) complexes for their potential antimicrobial activity against different strains of Klebsiella pneumoniae, Escherichia coli, and Pseudomonas aeruginosa which have developed different type of resistance mechanism, including the expression of β-lactamases (NDM-1, ESβL, or AmpC) or the production of biofilm. We studied a series of thiosemicarbabazones derived from pyridoxal, their related Ga(III) complexes, and the speciation in solution of the Ga(III)/ligand systems as a function of the pH. Proton dissociation constants and conditional stability constants of Ga(III) complexes were evaluated by UV/Vis spectroscopy, and the most relevant species at physiological pH were identified. The compounds are active against resistant Gram-negative strain with minimal inhibitory concentration in the μM range, while no cytotoxicity was detected in eukaryotic cells.

The use of dendrimers for metal cluster synthesis is an attractive approach for alloying multiple metal atoms into metal clusters because of the nanosized spaces and wide variety of metal salts that can be coordinated by dendrimers. However, the repeating structure of the dendrimers is generally not suitable for the precise assembly of different kinds of metal salts in them. Here, an amine/imine hybrid dendrimer was synthesized as a template for bimetallic assembly via the partial reduction of dendritic polyphenylazomethines (DPAs), which exhibit atomically precise metal salt assembly via 1:1 coordination between metal ions and their imine sites.

The molecular pathology of Alzheimer's disease is associated with the presence of aggregates of amyloid-β, a 39-43 amino acid peptide, that form amyloid plaques in the brain. Appropriately substituted stilbene derivatives, radiolabelled with positron-emitting radionuclides, that bind selectively to amyloid-β plaques can be used to assess plaque burden by Positron Emission Tomography (PET) imaging and assist in the diagnosis of Alzheimer's disease. In this work, a substituted pentadentate ligand based on a triazacyclononane backbone (HL) with one pendent stilbene functional group and two pendent carboxylate groups was synthesised.

Three 2-thiophenecarboxylate (Tio2c) complexes with different central atoms Ag(I), Zn(II) and Ga(III), [Ag(Tio2c)] (AgTio2c), {[Zn(Tio2c)]} (ZnTio2c) and [Ga(Tio2c)]·HO (GaTio2c), were synthesized and elemental, spectral and thermal analyses were used for their characterization. The AgTio2c and ZnTio2c single crystal structures confirmed the most common bidentate bridging coordination mode with typical strong argentophilic interactions in the case of AgTio2c complex. Complexes' stability in biological test stock solution were confirmed by H NMR spectroscopy.
