Article Abstract

β-Sheet forming polypeptides are one of the least explored synthetic systems due to their uncontrolled precipitation in the ring-opening polymerization (ROP) synthetic methodology. Here, a new -butylbenzene functionalization approach is introduced to overcome this limitation by sterically controlling the propagating polymer chains, and homogeneous polymerization with good control over chain growth was accomplished. New bulky -carboxyanhydride monomers were designed having -butylbenzene pendant by multistep organic synthesis, and N-heterocyclic carbene was explored as a catalyst to make high-molecular-weight and narrow polydisperse soluble polypeptides. This ROP process was successfully demonstrated for two β-sheet forming polypeptides such as poly(l-serine) and poly(l-cysteine). These new -butylbenzene-functionalized polypeptides were found to be readily soluble in tetrahydrofuran, chloroform, and so forth, and they were produced in high molecular weights having = 32 kDa with dispersity ≤ 1.3. ROP kinetics were studied by real-time Fourier transform infrared and H NMR to determine the actual content of the secondary structures in the propagating chains. These studies established that the α-helical conformational front in the propagation chain was speeding up the polymerization kinetics with good degree of control in the ROP process. Reversible-conformational transitions in the post-polymerization deprotection were found to restore the β-sheet secondary structures in poly(l-serine)s. The newly developed -butylbenzene-substituted steric-hindrance approach is valuable in yielding soluble polymers, and this approach could be useful for exploring new polypeptide architectures for long-term impact.

Background: Survivors of critical illness frequently face physical, cognitive and psychological impairments after intensive care. Sensorimotor impairments potentially have a negative impact on participation. However, comprehensive understanding of sensorimotor recovery and participation in survivors of critical illness is limited.

Experiencing stigmatization during the COVID-19 pandemic: a qualitative study among healthcare workers.

BMC Infect Dis

Department Health and Prevention, Institute of Psychology, University of Greifswald, Robert-Blum-Str. 13, Greifswald, 17489, Germany.

Background: Healthcare workers (HCWs) played a crucial role in dealing with the COVID-19 pandemic. In addition to increased workloads, they were confronted with stigmatization due to their work in the health sector.

Methods: Guided by the Health Stigma and Discrimination Framework (HSDF), this study aimed to explore the experiences of stigmatization of HCWs in Germany using semi-structured interviews (N = 34) and investigate effective coping strategies and existing needs in this context.

Large interstitial telomeric regions are considered remnants and markers of chromosomal rearrangements or a result of several suggested molecular mechanisms of telomere repeats accumulation. More rare are cases when large interstitial repeats are found not close to, but at a distance from the centromere. However, synapsis, recombination, and effects on chromatin near these regions during meiotic prophase I have not been sufficiently studied.
