Article Abstract

Compared to cell therapy, where cells are injected into a defect region, the treatment of heart infarction with cells seeded in a vascularized scaffold bears advantages, such as an immediate nutrient supply or a controllable and persistent localization of cells. For this purpose, decellularized native tissues are a preferable choice as they provide an in vivo-like microenvironment. However, the quality of such scaffolds strongly depends on the decellularization process. Therefore, two protocols based on sodium dodecyl sulfate or sodium deoxycholate were tailored and optimized for the decellularization of a porcine heart. The obtained scaffolds were tested for their applicability to generate vascularized cardiac patches. Decellularization with sodium dodecyl sulfate was found to be more suitable and resulted in scaffolds with a low amount of DNA, a highly preserved extracellular matrix composition, and structure shown by GAG quantification and immunohistochemistry. After seeding human endothelial cells into the vasculature, a coagulation assay demonstrated the functionality of the endothelial cells to minimize the clotting of blood. Human-induced pluripotent-stem-cell-derived cardiomyocytes in co-culture with fibroblasts and mesenchymal stem cells transferred the scaffold into a vascularized cardiac patch spontaneously contracting with a frequency of 25.61 ± 5.99 beats/min for over 16 weeks. The customized decellularization protocol based on sodium dodecyl sulfate renders a step towards a preclinical evaluation of the scaffolds.

Selective and rapid detection of ammonia (NH) gas over a wide concentration range is essential for applications such as early diagnosis of renal diseases and environmental safety. NH in exhaled breath serves as a biomarker of kidney function, and its precise detection is vital for early renal disease diagnosis. This work reports a SnS/PANI heterojunction nanocomposite (SPA) sensor synthesized a hydrothermal route followed by oxidative polymerization.

This study examined the pH-dependent (3, 5, and 7) regulation of matrix metalloproteinase (MMP) activity by cathepsin K (catK) and glycosaminoglycans (GAGs) using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), fluorescence assays, and human dentin slice experiments. The direct effects of catK were evaluated in the catK-active, catK-deficient, and odanacatib (ODN)-inhibited groups, whereas indirect GAG/ tissue inhibitor of metalloproteinase (TIMP)-mediated regulation was assessed in the catK-active, ODN-inhibited, and chondroitin sulfate (CS)-treated groups through dimethylmethylene blue (DMMB) assays, in situ zymography, and immunofluorescence staining. CatK directly activated MMP-2 (62 kDa) and MMP-9 (82 kDa) at all pH values, with no activation observed in the ODN-inhibited or catK-deficient groups.

The structure of amyloid-β (1-42) oligomers in membrane-mimetic environments.

Department of Biochemistry and Biophysics, Stockholm University, Sweden. Electronic address:

Aggregation of the amyloid-β peptide (Aβ) characterises and probably causes Alzheimer's disease. While lipid-mediated Aβ aggregation has been extensively studied for the 40-residue variant Aβ40, the interaction of the 42-residue variant Aβ42 with membranes has received less attention. Our time-resolved infrared spectra demonstrate that Aβ42 oligomers preserve their β-sheet structure in aqueous solution also in a membrane-mimicking environment consisting of either 1-hexadecanoyl-2-(9Z-octadecenoyl)-sn-glycero-3-phosphocholine (POPC, zwitterionic) or 1-hexadecanoyl-2-(9Z-octadecenoyl)-sn-glycero-3-phospho-(1'racglycerol) (POPG, anionic) vesicles.

A purified-keratin solution obtained from wool fibers by sulfitolysis extraction was employed to produce hydrogels with and without crosslinking. Both hydrogels were used to successfully obtain aerogels by supercritical CO drying. Freeze-dried keratin was also produced from purified keratin solutions as reference materials.
