qPCR absolute quantification requires the assessment of PCR reaction efficiency. This is achieved by performing serial dilutions of gDNA. Herein, we demonstrate that when quantifying bacterial load in mixed samples, reaction efficiency should not be used as a calibration curve since gDNA isolation efficiency is neglected, significantly impacting quantification.

Background: Transforming Clinical Practice Guideline (CPG) recommendations into computer readable language is a complex and ongoing process that requires significant resources, including time, expertise, and funds. The objective is to provide an extension of the widely used GIN-McMaster Guideline Development Checklist (GDC) and Tool for the development of computable guidelines (CGs).
Methods: Based on an outcome from the Human Centered Design (HCD) workshop hosted by the Guidelines International Network North America (GIN-NA), a team was formed to develop the checklist extension.

Neutrophil extracellular traps (NETs) are DNA-protein structures released during a form of programmed neutrophil death known as NETosis. While NETs have been implicated in both tumor inhibition and promotion, their functional role in cancer remains ambiguous. In this study, we compared the NET-forming capacity and functional effects of NETs derived from lung cancer (LC) patients and healthy donors (H).

Background: Revision hip arthroplasty is associated with significant blood loss and a subsequent need for blood transfusion. Intraoperative cell salvage (ICS) is a method of recovering a patient's blood for autologous transfusion. This potentially reduces the need for allogenic blood transfusion (ABT), thus avoiding associated risk and expense.

Background: Acid sphingomyelinase deficiency (ASMD) type A/B, a rare lysosomal storage disorder caused by biallelic mutations in the SMPD1 gene, presents with variable visceral and neurological manifestations. Arnold-Chiari malformation is a structural defect of the cerebellum and brainstem with distinct pathogenesis and clinical course. To our knowledge, the coexistence of these two conditions has not been previously reported.
