Aquaporins facilitate the passive transport of water, solutes, or ions across biological membranes. They are implicated in diverse pathologies including brain edema following stroke or trauma, epilepsy, cancer cell migration and tumor angiogenesis, metabolic disorders, and inflammation. Despite this, there is no aquaporin-targeted drug in the clinic and aquaporins have been perceived to be intrinsically non-druggable targets. Here we challenge this idea, as viable routes to inhibition of aquaporin function have recently been identified, including targeting their regulation or their roles as channels for unexpected substrates. Identifying new drug development frameworks for conditions associated with disrupted water and solute homeostasis will meet the urgent, unmet clinical need of millions of patients for whom no pharmacological interventions are available.

The landscape of allosteric drug discovery is undergoing a transformative shift, driven by the integration of three computational approaches: machine learning (ML), molecular dynamics (MD) simulations, and network theory. ML identifies potential allosteric sites from multidimensional biological datasets; MD simulations, empowered by enhanced sampling algorithms, reveal transient conformational states; and network analyses uncover communication pathways, further aiding in site identification. Their synergy enables rational allosteric modulator design.

The coastal mixing zone between seawater and freshwater is a critical interface for the exchange and transformation of contaminants. Amorphous materials with thickness thinned down to the single-layer limit have attracted increasing interest due to their well-defined disorder and emerging unique properties, such as disorder-dominated electronic states, high-density unsaturated coordination, enhanced quantum confinement, These features could enable innovative applications in electronics, photoelectronics, catalysis, and beyond. In this perspective, we provide an overview of recent advances in two-dimensional (2D) amorphous materials approaching the single-layer limit. We first introduce newly-developed key structural descriptors for these systems, including local bonding, topological disorder, and chemical composition.
