A novel mRNA-based multi-epitope vaccine for rabies virus computationally designed via reverse vaccinology and immunoinformatics.

Sci Rep

August 2025

Research laboratory for Epidemiology and immunogenetics of viral infections (LR14SP02), Sahloul University Hospital, University of Sousse, Sousse, Tunisia.

The current research investigated the development of a multi-epitope mRNA vaccine against the rabies virus on the basis of viral proteomes via the use of bioinformatic tools and reverse vaccinology. The aim of this study was to address the limitations of the currently available rabies vaccine by eliciting strong and long-lasting humoral and cellular immune responses. The cytotoxic T lymphocytes (CTLs), helper T lymphocytes (HTLs), and linear B-cell epitopes (LBLs) were mapped and prioritized from four top-ranking vaccine targets (nucleoprotein, phosphoprotein, matrix, and glycoprotein) that were highly antigenic, nonallergenic, nontoxic, and nonhuman homologs.

It has been recognized that it is difficult to maintain the virus particle structure of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which may be associated with lower immunogenicity of inactivated vaccines against coronavirus disease 2019 (COVID-19). We have previously demonstrated that an intact structure of the virus particles is critical for influenza inactivated whole virus particle vaccine (WPV) to be immunogenically potent. Here, we tested 37, 35, 33, and 31 °C for the virus propagation temperatures and the timing of formaldehyde treatment of the virus before and after centrifugation-based purification to obtain virus particles with an intact structure.

The formation of antidrug antibodies (ADAs) against antibody-drug conjugates (ADCs) can trigger a humoral immune response and change drug exposure. Although the immunogenicity assessment of an ADC drug in nonclinical nonhuman primates may not directly translate to potential immunogenicity in humans, the nonclinical ADA assay facilitates understanding the pharmacokinetic profiles of biotherapeutics. The immune response against the human IgG4 monoclonal antibody-based ADC was suspected in cynomolgus monkey serum after intravenous administration at 1.

Immunological interventions, like vaccinations, are enabled by the predictive control of humoral responses to novel antigens. While the development trajectories for many broadly neutralizing antibodies (bnAbs) have been measured, it is less established how human subtype-specific antibodies develop from their precursors. In this work, we evaluated the retrospective development trajectories for eight anti-SARS-CoV-2 Spike human antibodies (Abs).

Dental caries, a persistent oral health challenge primarily linked to , extends its implications beyond dental decay, affecting over 4 billion individuals globally. Despite its historical association with childhood, dental caries often persists into adulthood with prevalence rates ranging from 60 to 90% in children and 26 to 85% in adults. Currently, there is a dearth of multiepitope vaccines (MEVs) specifically designed to combat .
