Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the recently emerged virus responsible for the COVID-19 pandemic. Clinical presentation can range from asymptomatic disease and mild respiratory tract infection to severe disease with lung injury, multiorgan failure, and death. SARS-CoV-2 is the third animal coronavirus to emerge in humans in the 21st century, and coronaviruses appear to possess a unique ability to cross borders between species and infect a wide range of organisms. This is somewhat surprising as, except for the requirement of host cell receptors, cell-pathogen interactions are usually species-specific. Insights into these host-virus interactions will provide a deeper understanding of the process of SARS-CoV-2 infection and provide a means for the design and development of antiviral agents. In this study, we describe a complex analysis of SARS-CoV-2 infection using a genome-wide CRISPR-Cas9 knock-out system in HeLa cells overexpressing entry receptor angiotensin-converting enzyme 2 (ACE2). This platform allows for the identification of factors required for viral replication. This study was designed to include a high number of replicates (48 replicates; 16 biological repeats with 3 technical replicates each) to prevent data instability, remove sources of bias, and allow multifactorial bioinformatic analyses in order to study the resulting interaction network. The results obtained provide an interesting insight into the replication mechanisms of SARS-CoV-2.

To analyze in-hospital mortality in children undergoing congenital heart interventions in the only public referral center in Amazonas, North Brazil, between 2014 and 2022. This retrospective cohort study included 1041 patients undergoing cardiac interventions for congenital heart disease, of whom 135 died during hospitalization. Records were reviewed to obtain demographic, clinical, and surgical data.

Purpose Of Review: This review aims to characterize the known cardiovascular (CV) manifestations associated with inflammatory bowel disease (IBD) and the underlying mechanisms driving these associations.
Recent Findings: Gut dysbiosis, a hallmark of patients with IBD, can result in both local and systemic inflammation, thereby potentially increasing the risk of cardiovascular disease (CVD) in the IBD population. Micronutrient deficiencies, anemia, and sarcopenia independently increase the risk of CVD and are frequent comorbidities of patients with IBD.

Purpose: Sleep disturbance is prevalent in long-term care facilities (LTCFs), yet there is limited understanding of individual factors predicting changes in sleep within these populations. Our objective was to determine predictors of sleep disturbance in LTCFs and investigate variation in prevalence across facilities in two Canadian provinces-New Brunswick and Saskatchewan.
Method: This retrospective longitudinal cohort study used interRAI comprehensive health assessment data from 2016 to 2021, encompassing 21,394 older adults aged ≥ 65 years across 228 LTCFs.

Purpose: Antibiotic-sparing treatment (ASPT) strategies, such as delayed prescribing and symptomatic treatment, are promising to reduce antimicrobial consumption (AMC) in patients with uncomplicated urinary tract infections (uUTI). The aim of this scoping review was to identify literature reporting on factors that may act as barriers and facilitators to the use of ASPT in order to improve implementation.
Methods: MEDLINE (Ovid), Embase, the Cochrane Database, Google Scholar, Proquest Dissertations and Theses, the Clinical Trials Gov Registry and the ICTRP WHO Registry were searched for evidence of health care professionals and/or patients exposed to ASPT in the context of uUTI.
