Tetrazoles were designed and synthesized as potential inhibitors of triple monoamine neurotransmitters (dopamine, norepinephrine, serotonin) reuptake based on the functional and docking simulation of compound which were performed in a previous study. The compound structure consisted of a tetrazole-linker (n)-piperidine/piperazine-spacer (m)-phenyl ring, with tetrazole attached to two phenyl rings (R1 and R2). Altering the carbon number in the linker (n) from 3 to 4 and in the spacer (m) from 0 to 1 increased the potency of serotonin reuptake inhibition. Depending on the nature of piperidine/piperazine, the substituents at R1 and R2 exerted various effects in determining their inhibitory effects on monoamine reuptake. Docking study showed that the selectivity of tetrazole for different transporters was determined based on multiple interactions with various residues on transporters, including hydrophobic residues on transmembrane domains 1, 3, 6, and 8. Co-expression of dopamine transporter, which lowers dopamine concentration in the biophase by uptaking dopamine into the cells, inhibited the dopamine-induced endoctytosis of dopamine D receptor. When tested for compound and , compound which has more potent inhibitory activity on dopamine reuptake more strongly disinhibited the inhibitory activity of dopamine transporter on the endocytosis of dopamine D receptor. Overall, we identified candidate inhibitors of triple monoamine neurotransmitter reuptake and provided a theoretical background for identifying such neurotransmitter modifiers for developing novel therapeutic agents of various neuropsychiatric disorders.

Background: Vitamin D, sometimes referred to as the "sunshine vitamin", is well-known for its role in maintaining bone health processes that are highly dependent on calcium regulation. However, there is an emerging wealth of evidence that this fat-soluble vitamin has an important role in male sexual health regarding erectile function, production of testosterone, and overall fertility. Due to the increased vitamin D deficiency rates in most populations, the implications of its deficiency on male sexual functions have gained great interest.

Ketamine has been widely used as a recreational substance by adolescents and young adults in nightclubs and raves in an acute manner, especially during the weekend. Considering the scarcity of evidence on the harmful consequences of adolescent ketamine recreational use on the central nervous system, primarily related to motor function, this study aimed to investigate the behavioral, biochemical, and neurochemical consequences on motor function induced by ketamine use, evaluating the motor cortex, cerebellum, and striatum in early abstinence. Adolescent female Wistar rats (28 days old) received ketamine by intranasal route (10mg/kg/day) for 3 consecutive days.

The transition from preclinical to clinical drug development is critically impeded by interspecies disparities, which limit the predictive validity of preclinical efficacy for human outcomes. To address this limitation, we established a human flora-associated depression rat (HFADR) model through fecal microbiota transplantation (FMT). The HFADR model bridges the preclinical-clinical translation by recapitulating conserved microbial-host interactions identified through multi-omics analysis in a chronic unpredictable mild stress (CUMS) rat model and in patients with major depressive disorder.

Background: Sleep disorders show comorbidity with depression and Alzheimer's disease (AD), especially in ageing. However, the neuroimmunological role of sleep deprivation (SD) as possible inducer to these conditions remains unknown. Omega-3 fatty acids (n-3 FAs) can improve depression and AD through anti-inflammation, up-regulating neurotrophins and normalizing neurotransmitters, while their therapeutic effects on sleep deprivation (SD)-induced changes in different ages requires investigation.

It is a promising dietary supplement for the treatment of depression based on TCM principles and nutritional assessments. Our study was designed to explore BCBL's antidepressant-like effects and elucidate its underlying mechanisms.
