Persistent pain affecting patients with inflammatory bowel diseases (IBDs) is still very difficult to treat. Carbonic anhydrase (CA) represents an intriguing pharmacological target considering the anti-hyperalgesic efficacy displayed by CA inhibitors in both inflammatory and neuropathic pain models. The aim of this work was to evaluate the effect of inhibiting CA IV, particularly when expressed in the gut, on visceral pain associated with colitis induced by 2,4-di-nitrobenzene sulfonic acid (DNBS) in rats. Visceral sensitivity was assessed by measuring animals' abdominal responses to colorectal distension. Repeated treatment with the selective CA IV inhibitors AB-118 and NIK-67 effectively counteracted the development of visceral pain induced by DNBS. In addition to pain relief, AB-118 showed a protective effect against colon damage. By contrast, the anti-hyperalgesic activity of NIK-67 was independent of colon healing, suggesting a direct protective effect of NIK-67 on visceral sensitivity. The enzymatic activity and the expression of CA IV resulted significantly increased after DNBS injection. NIK-67 normalised CA IV activity in DNBS animals, while AB-118 was partially effective. None of these compounds influenced CA IV expression through the colon. Although further investigations are needed to study the underlying mechanisms, CA IV inhibitors are promising candidates in the search for therapies to relieve visceral pain in IBDs.

The connective tissue support of female pelvic viscera-endopelvic fascia-has been studied in fetal and immunohistochemical models to demonstrate its relationship with the autonomic nerves of the female pelvis. Due to a paucity of literature examining the gross anatomical relationships between endopelvic fascia and autonomic nerves in adult female pelvises, it remains unknown whether defects in endopelvic fascia predisposing pelvic organ prolapse and/or manipulation of endopelvic fascia during prolapse repair may be the cause of prolapse-related pelvic pain and sexual dysfunction. Through the dissection of formalin-fixed hemipelvises (n = 10) the present study aimed to map the loci of the visceral branches of the inferior hypogastric plexus and associate them with endopelvic fascia of the female pelvis.

Objective: To investigate the prevalence of mesenteric artery stenosis and its association with acute mesenteric ischemia (AMI) among elderly patients presenting at the emergency department with acute abdominal pain.
Methods: This single-center retrospective cohort study included 500 consecutive patients aged 65 years or more who underwent contrast enhanced computed tomography at the emergency department due to acute abdominal pain between 2013 and 2014. Imaging data were retrospectively evaluated by a consultant interventional radiologist for ≥50% stenosis of the superior mesenteric artery (SMA), celiac artery (CA) and inferior mesenteric artery (IMA).

Chronic gastrointestinal pain is a hallmark of most intestinal pathologies, yet effective treatments remain elusive given the complexity of the underlying mechanisms. Aiming to investigate the intestinal epithelium contribution to visceral pain modulation in dysbiosis context, we first demonstrated that intracolonic instillation of microbe-free fecal supernatants from mice with post-inflammatory dysbiosis induced by dextran sodium sulfate (FS) provokes visceral hypersensitivity in recipient mice. Epithelium involvement in the response to FS was analyzed through a novel approach comprising murine epithelial colon organoids and primary dorsal root ganglia (DRG) neurons.

Formaldehyde (commonly in the form of formalin solution) is widely used as tissue fixative in clinics and laboratories. If inadvertently introduced into living tissue, formaldehyde causes rapid protein coagulation and tissue fixation, leading to cell death and necrosis.
