Article Abstract

We have performed fully atomistic molecular dynamics simulations of the intracellular domain of a model of the GABA receptor with and without the GABA receptor associated protein (GABARAP) bound. We have also calculated the electrostatic potential due to the receptor, in the absence and presence of GABARAP. We find that GABARAP binding changes the electrostatic properties around the GABA receptor and could lead to increased conductivity of chloride ions through the receptor. We also find that ion motions that would result in conducting currents are observed nearly twice as often when GABARAP binds. These results are consistent with data from electrophysiological experiments.

Although glutamatergic and GABAergic synapses are important in seizure generation, the contribution of non-synaptic ionic and electrical mechanisms to synchronization of seizure-prone hippocampal neurons remains unclear. Here, we developed a physiologically relevant model to study these mechanisms by inducing prolonged seizure-like discharges (SLDs) in hippocampal slices from male rats through modest, sustained ionic manipulations. Specifically, we reduced extracellular calcium to 0.

Autism spectrum disorders (ASD), a group of neurodevelopmental disorders characterized by the core symptoms of impaired social communication and stereotyped behaviors, is strongly associated with dysregulated microbiota-gut-brain axis. Emerging evidence suggests that , which showed reduced abundance in ASD cohorts, holds therapeutic potential, though its interaction with host remain unexplored. Here, we investigated the efficacy and molecular basis of 4P-15 (4P-15) in BTBR /J (BTBR) mice, an idiopathic ASD mouse model.

Luzhou Key Laboratory of Traditional Chinese Medicine for Chronic Diseases Jointly Built by Sichuan and Chongqing, The Affiliated Traditional Chinese Medicine Hospital, Southwest Medical University, Luzhou, Sichuan, China.

Introduction: Depression and anxiety are prevalent comorbidities in individuals with chronic diseases, significantly impairing their quality of life and complicating disease management. Curcumin, derived from turmeric (Curcuma longa), has garnered attention for its potential therapeutic benefits in alleviating symptoms of depression and anxiety. However, its specific effects on depressive or anxiety symptoms associated with chronic diseases (DACD) remain unclear.

Introduction: Neuropathic pain severely affects patients' quality of life. Limited treatments offer relief but often involve long-term use and side effects.

Objectives: This study aimed to explore the electroacupuncture (EA) and subanesthetic alfaxalone (ALF) combination as a novel therapeutic substitute for the treatment of neuropathic pain in a spared nerve injury (SNI) mouse model.

Department of Nursing, Shenzhen Hospital, Southern Medical University, Shenzhen 518101, China.

Objectives: To investigate the therapeutic effect of acupuncture in a rat model of insomnia and its regulatory effect on the glutamic acid (Glu)/γ-aminobutyric acid (GABA)-glutamine (Gln) metabolic loop.

Methods: Forty male SD rats were randomly assigned to control group, model group, group and group (=10). In the latter 3 groups, rat models of insomnia were established by intraperitoneal injections of p-chlorophenylalanine and verified using a sodium pentobarbital-induced sleep test.
