Article Abstract

Escape from threats has paramount importance for survival. However, it is unknown if a single circuit controls escape vigor from innate and conditioned threats. Cholecystokinin (cck)-expressing cells in the hypothalamic dorsal premammillary nucleus (PMd) are necessary for initiating escape from innate threats via a projection to the dorsolateral periaqueductal gray (dlPAG). We now show that in mice PMd-cck cells are activated during escape, but not other defensive behaviors. PMd-cck ensemble activity can also predict future escape. Furthermore, PMd inhibition decreases escape speed from both innate and conditioned threats. Inhibition of the PMd-cck projection to the dlPAG also decreased escape speed. Intriguingly, PMd-cck and dlPAG activity in mice showed higher mutual information during exposure to innate and conditioned threats. In parallel, human functional magnetic resonance imaging data show that a posterior hypothalamic-to-dlPAG pathway increased activity during exposure to aversive images, indicating that a similar pathway may possibly have a related role in humans. Our data identify the PMd-dlPAG circuit as a central node, controlling escape vigor elicited by both innate and conditioned threats.

Adjuvants are widely used to boost the immune response during vaccination protocols. Our group has previously reported that repeated intraperitoneal administration of alum in mice, known as adjuvant conditioning (AC), creates an immunosuppressive environment that delays allogeneic graft rejection through NLRP3-dependent MDSC expansion. Innate behaviours allow solitary animals to complete essential tasks in the absence of social learning. However, we know little about the degree to which ecologically relevant innate preferences can change, and a complete extinction of innate preferences has rarely been shown. The hoverfly Eristalis tenax, a solitary generalist pollinator, is an ideal model for studying innate behaviour in a naturalistic context because its survival depends on the innate ability to identify flowers across many habitats, which could necessitate both learning and unlearning floral objects.

Introduction: Mesenchymal stromal cells (MSCs) have been evaluated as a local therapeutic option to treat osteoarthritis (OA) with conflicting clinical results. Our previous studies have evaluated immune licensing of MSC through activation of Toll-like receptor and cytosolic cGAS-STING pathways, with demonstrated improvement in functional and structural outcomes in a rodent model of OA. The objective of this study was to investigate impact of MSC activation on their interaction with relevant joint target cells to better understand the mechanisms by which pre-activation improves MSC activity for treatment of osteoarthritis.

Aversion modulation is a key component of hedonic processing, and its dysfunction is evident in psychiatric illnesses. The infralimbic cortex (IL) to nucleus accumbens shell (NAcSh) pathway is essential in hedonic processing in rodents but operates differentially across sex, with beta (20 Hz) oscillatory activity involved in learned aversion in male but not female rats. In this study, we used taste reactivity (TR) and electrophysiology to examine the role of high gamma (80 Hz) activity in affect modulation, specifically innate (quinine) and learned (conditioned taste aversion, CTA) aversion, in male and female Sprague-Dawley rats.
