Oligodendrocytes are highly specialized glial cells, responsible for producing myelin in the central nervous system (CNS). The multi-stage process of oligodendrocyte development is tightly regulated to ensure proper lineage progression of oligodendrocyte progenitor cells (OPCs) to mature myelin producing oligodendrocytes. This developmental process involves complex interactions between several intrinsic signaling pathways that are modulated by an array of extrinsic factors. Understanding these regulatory processes is of crucial importance, as it may help to identify specific molecular targets both to enhance plasticity in the normal CNS and to promote endogenous recovery following injury or disease. This review describes two major regulators that play important functional roles in distinct phases of oligodendrocyte development: OPC proliferation and differentiation. Specifically, we highlight the roles of the extracellular astrocyte/radial glia-derived protein Endothelin-1 in OPC proliferation and the intracellular Akt/mTOR pathway in OPC differentiation. Lastly, we reflect on how recent advances in neuroscience and scientific technology will enable greater understanding into how intrinsic and extrinsic regulators interact to generate oligodendrocyte diversity.

The wrist extends anatomically and functionally from the distal forearm to the metacarpal bones and encompasses the entire carpal region. The large number of bones and joints involved are stabilized in a highly complex system of extrinsic and intrinsic ligaments, which enables a variety of different movements and stability in all positions for the best possible use of the hands. While injuries to individual ligaments or joints, most commonly the scapholunate (SL) ligament, can cause major problems the injuries associated with perilunate dislocations and dislocated fractures represent one of the greatest challenges in hand surgery.

Small antral follicles frequently undergo atresia due to inadequate gonadotropin support, characterized by reduced estrogen synthesis and granulosa cell (GC) apoptosis. Integrins bind ligands between their alpha (α) and beta (β) subunits and transmit signals through conformational changes. Early in chordate evolution, some α subunits acquired an "inserted" (I) domain that expanded integrin's ligand-binding repertoire but obstructed the ancestral ligand pocket, seemingly blocking conventional integrin activation. Here, we compare cryo-electron microscopy structures of apo and ligand-bound states of the I domain-containing αEβ integrin and the I domain-lacking αβ integrin to illuminate how the I domain intrinsically mimics an extrinsic ligand to preserve integrin function.

Bone stress injury is a common musculoskeletal condition presenting with insidious bony pain that is progressive and occurs with a number of intrinsic or extrinsic risk factors, particularly with a recent change in training. When elicited, the presence of bony tenderness remains the most important component of the physical exam, although reproduction at deeper sites is a challenge and requires a high index of suspicion and imaging for diagnosis. MRI should be utilized as the gold standard for diagnosis, grading, and return-to-sport timing prognosis when available, with plain radiographs used as first-line imaging.
