Article Abstract

This work reviews the literature of chiral capillary electrokinetic chromatography from January 2016 to March 2021. This is done to explore the state-of-the-art approach and recent developments carried out in this field. The separation principle of the technique is described and supported with simple graphical illustrations, showing migration under normal and reversed polarity modes of the separation voltage. The most relevant applications of the technique for enantioseparation of drugs and other enantiomeric molecules in different fields using chiral selectors in single, dual, or multiple systems are highlighted. Measures to improve the detection sensitivity of chiral capillary electrokinetic chromatography with UV detector are discussed, and the alternative aspects are explored, besides special emphases to hyphenation compatibility to mass spectrometry. Partial filling and counter migration techniques are described. Indirect identification of the separated enantiomers and the determination of enantiomeric migration order are mentioned. The application of Quality by Design principles to facilitate method development, optimization, and validation is presented. The elucidation and explanation of chiral recognition in molecular bases are discussed with special focus on the role of molecular modeling.

The enantiomeric distribution of the δ- and γ-lactones in Wagyu beef was determined using solvent extraction and gas chromatography with a chiral capillary column. The results revealed that the predominant lactones in Wagyu were (R)-δ-tetradecalactone and (R)-δ-hexadecalactone. The concentrations of several lactone types were higher after heat treatment.

The development of novel enantioseparation materials requires an effective testing method that not only is user-friendly, rapid, and reproducible but also minimizes the amount of enantioseparation material needed. In this work, commercially available silica particles coated with cellulose tris(3,5-dimethylphenylcarbamate) were employed as the chiral stationary phase to develop a simple and rapid method for preparing chiral packed capillary columns compatible with conventional CEC-UV or cLC-UV systems. The column fabrication process took only 2 h.

Therapeutic peptides have gained prominence in the pharmaceutical market, but among other reasons, their development was hindered by the risk of amino acid epimerization, particularly histidine (His), which is prone to alter its chiral configuration (L or D) during synthesis. Covalent organic frameworks (COFs), with high surface area, porous structure, and biocompatibility, offer an ideal platform for enzyme immobilization.
