Article Abstract

3D tumor spheroids have arisen in the last years as potent tools for the in vitro screening of novel anticancer therapeutics. Nevertheless, to increase the reproducibility and predictability of the data originated from the spheroids it is still necessary to develop or optimize the techniques used for spheroids' physical and biomolecular characterization. Fluorescence microscopy, such as confocal laser scanning microscopy (CLSM), is a tool commonly used by researchers to characterize spheroids structure and the antitumoral effect of novel therapeutics. However, its application in spheroids' analysis is hindered by the limited light penetration in thick samples. For this purpose, optical clearing solutions have been explored to increase the spheroids' transparency by reducing the light scattering. In this study, the influence of agitation conditions (i.e., static, horizontal agitation, and rotatory agitation) on the and methods' clearing efficacy and tumor spheroids' imaging by CLSM was characterized. The obtained results demonstrate that the method results in the improved imaging of the spheroids interior, whereas the resulted in an increased propidium iodide mean fluorescence intensity as well as a higher signal depth in the Z-axis. Additionally, for both methods, the best clearing results were obtained for the spheroids treated under the rotatory agitation. In general, this work provides new insights on the and clearing methodologies and their utilization for improving the reproducibility of the data obtained through the CLSM, such as the analysis of the cell death in response to therapeutics administration.

With the acceleration of global industrialization, a large amount of polluted wastewater is discharged indiscriminately, which both pollutes the environment and threatens human health. In this study, by constructing a binary system of unsaturated polyester resin/carboxychitosan, and improving the inherent defects of carboxychitosan aerogel, we successfully prepared aerogels with high porosity, low density, and laminar porous structure for water remediation by using a combination of the sol-gel method and directional freezing technology. Thanks to the synergistic effect of surface wettability and special pore structure, the aerogel not only adsorbs and separates MB and Pb(II) efficiently with a separation efficiency of more than 99 %, but also has a separation efficiency of 99.

The perception of olfactory stimuli can influence animal emotions and behaviours, which are critical for survival and adaptation. This study explored the properties of three olfactory stimuli (orange essential oil, wolf faeces, and cadaverine) to induce emotion in sheep, in a non-stressful environment enriched with social and food resources. Contrary to expectations, wolf faeces, while repellent, did not trigger overt stress behaviours such as increased vocalisations or agitation.

Endotoxins are components of the outer membrane of bacteria that can become airborne during aerosol-generating work activities and cause adverse effects on workers' health. Filtration is the sampling method recommended by the EN 14031 standard for endotoxin measurements in workplace atmospheres.
