Article Abstract

An atom-economic Pd(ii)-catalysed cascade cyclisation of 2-(biphenylethynyl)anilines tethered to an aldehyde or cyano group leads to the formation of dibenzo[5,6:7,8]cycloocta[1,2-b]indol-10-ols 6 or dibenzo[5,6:7,8]cycloocta[1,2-b]indol-10(15H)-ones 8 with high yields (up to 95%). The reaction proceeds via amino-palladation of the alkyne followed by nucleophilic addition onto the aldehyde/cyano group. Treatment of 6 with p-TsOH·HO smoothly provided cyclooctatetraene (COT) derivatives 7.

ConspectusFunctionalization of carbon-hydrogen (C-H) bonds has emerged as a powerful strategy in modern organic synthesis, offering efficient routes to build molecular complexity from simple and abundant substrates. Among various transition-metal catalysts, palladium(II) complexes have proven particularly versatile for C-H activation, owing to the diverse reactivity of carbon-palladium bonds. To advance this approach, the discovery of ligands that can accelerate C-H activation as well as subsequent steps in the catalytic cycle is the pivotal driving force.

-alkyl arylamines are important structural motifs in pharmaceuticals, yet traditional alkylating methods rely on the nucleophilicity of the amine and make access to such compounds with valuable bioproperties challenging. While metal-mediated reactions may alleviate these limitations, they often encounter amine-metal interactions that can hinder catalysis or lead to deleterious pathways. Herein, we report a palladium(II) [Pd(II)]/sulfoxide-oxazoline(SOX)/phosphoric acid-mediated C(sp)H/N(sp) cross-coupling of 53 arylamine nucleophiles and 39 terminal olefins to furnish >80 diverse tertiary (3°) arylamines in excellent yields (average 82%) and selectivities (>20:1 /, >20:1 linear/branched).

The mechanism by which Pd precursors are reduced to catalytically active low-valent Pd species has been a subject of interest for developing better catalysts. This process is well understood for catalytic systems employing a combination of palladium(ii) acetate [Pd(OAc)] and tertiary phosphines. However, the mechanism of reduction of palladium(ii) acetylacetonate [Pd(acac)] in the presence of phosphines has not been thoroughly investigated.

Organosulfur/selenium embedded heterocycles are high-value, intricate synthetic targets. Their cogent synthesis and postsynthetic diversifications, particularly through the C─H bond activation strategy, remain immensely alluring in organic synthesis. In this study, we illustrate the first example of site-selective C(sp)2-H chalcogenation (sulfenylation) of 4-quinolone motifs under palladium(II) catalysis in conjunction with earth-abundant copper salt as an oxidizing agent, enabling structurally orchestrated C-2 sulfenylated 4-quinolone scaffolds in promising yields.

Phosphine-free catalysts for the Mizoroki-Heck cross-coupling reaction offer significant advantages in terms of air and moisture stability. We synthesized two palladium(II) complexes featuring a thiolate-imine-thioether SNS pincer ligand: Pd(κ-SNS) (Pd-1) and PdI(κ-SNS) (Pd-2). Their catalytic activities were compared, with Pd-2 demonstrating excellent reactivity at a low catalyst loading of 1.
