Innate lymphoid cells (ILCs) develop from common lymphoid progenitors (CLPs), which further differentiate into the common ILC progenitor (CILP) that can give rise to both ILCs and natural killer (NK) cells. Murine ILC intermediates have recently been characterized, but the human counterparts and their developmental trajectories have not yet been identified, largely due to the lack of homologous surface receptors in both organisms. Here, we show that human CILPs (CD34CD117α4β7Lin) acquire CD48 and CD52, which define NK progenitors (NKPs) and ILC precursors (ILCPs). Two distinct NK cell subsets were generated in vitro from CD34CD117α4β7LinCD48CD52 and CD34CD117α4β7LinCD48CD52 NKPs, respectively. Independent of NKPs, ILCPs exist in the CD34CD117α4β7LinCD48CD52 subset and give rise to ILC1s, ILC2s, and NCR ILC3s, whereas CD34CD117α4β7LinCD48CD52 ILCPs give rise to a distinct subset of ILC3s that have lymphoid tissue inducer (LTi)-like properties. In addition, CD48-expressing CD34CD117α4β7Lin precursors give rise to tissue-associated ILCs in vivo. We also observed that the interaction of 2B4 with CD48 induced differentiation of ILC2s, and together, these findings show that expression of CD48 by human ILCPs modulates ILC differentiation.

Communication between group 3 innate lymphoid cells (ILC3) and other immune cells, as well as intestinal epithelial cells, is pivotal in regulating intestinal inflammation. This study, for the first time, underscores the importance of crosstalk between intestinal endothelial cells (ECs) and ILC3. Our single-cell transcriptome analysis combined with protein expression detection revealed that ECs significantly increased the population of interleukin (IL)-22 ILC3 through interactions mediated by endothelin-1 (ET-1) and its receptor endothelin A receptor (EDNRA).

Background: Breast cancer is a systemic disease, yet the impact of tumor molecular subtype and disease stage on the systemic immune landscape remains poorly understood. In this study, we comprehensively analyzed the systemic immune landscape in a large cohort of breast cancer patients, encompassing all molecular subtypes and disease stages, alongside a control group of healthy donors.
Materials And Methods: Using multi-parameter flow cytometry, we assessed the abundance, phenotype, and activation status of diverse innate and adaptive immune cell populations across peripheral blood samples from 355 breast cancer patients and 65 healthy donors.

Introduction: Although Coeliac disease (CD) current and only treatment is a life-long strict gluten free diet (GFD), some patients suffer from persistent duodenal lesions despite years into the diet. Hence, we aimed to study the effect that the GFD elicits on the mucosal immune infiltrate from these patients.
Method: To that end, duodenal biopsies were collected from non-coeliac controls and CD patients, both at diagnosis and after at least one year into the GFD.

The fraction that the elderly represent in the world's population is growing rapidly; numerous alterations that impact all organs and systems, including the immune system, are related to aging. A complex process common in the elderly, known as immunosenescence, is characterized by a decreased ability to respond to vaccination as well as an increased risk of bacterial and viral infections, autoimmune, cardiovascular and neurodegenerative diseases. These processes are associated with alterations in the innate and adaptive immune system and lead to a condition of chronic low-grade inflammation, referred to as inflammaging.
