Catchment-scale effects of river fragmentation: A case study on restoring connectivity.

Article Abstract

More than two thirds of large rivers worldwide are fragmented, threatening freshwater biodiversity, river integrity, and the services that freshwater ecosystems provide for human populations around the globe. In an effort to alleviate the impacts of barriers, engineered solutions have been developed, though with somewhat underwhelming results. River restoration, especially dam removal, is viewed as the optimal option though seldom the go-to approach. In this study, we evaluated the effects of a large restoration project (pseudo dam removal) in River Kolding, Southern Jutland, Denmark, via a before-after-control-impact (BACI) approach. Using a large dataset of electrofishing data from 74 sites (including downstream unaffected sites, reconnected sites and upstream regulated sites), we found that habitat connectivity was restored successfully, with a large increase in young-of-the-year brown trout (Salmo trutta) at reconnected sites, reaching similar densities to downstream (non-affected) sites. We further observed a decrease in length at reconnected sites, suggesting that natural spawning and rearing habitats were successfully restored too.

Breast cancer is a leading cause of death among women globally, with triple-negative breast cancer (TNBC) being the most aggressive subtype. To develop effective and safe chemotherapeutic drugs, we identified TOP2A as a potential target due to its differential expression between breast cancer and normal tissues. By targeting hydrogen bond-van der Waals sites, we discovered three active amino acid sites in TOP2A (E461, D463, and D543) and designed a β-carboline derivative, DM1, targeting DNA and TOP2A using a molecular generation model, local docking, and optimization.

The turnover and re-establishment of peripheral taste synapses is vital to maintain connectivity between primary taste receptor cells and the gustatory neurons which relay taste information from the tongue to the brain. Despite the importance of neuron-taste cell reconnection, the mechanisms governing synapse assembly in the taste bud are largely unknown. To determine whether nerve fiber connectivity is an initiating factor for the recruitment of presynaptic machinery in taste receptor cells, we use the expression of CALHM1 and Bassoon to identify presynaptic sites in type II (sweet, umami, bitter) and type III (sour) taste receptor cells, respectively.

Restoring connectivity via assisted migration is a useful but currently underused approach for maintaining genetic diversity and preventing extirpations of threatened species. The use of assisted migration as a conservation strategy may be limited by the difficulty of balancing the benefits of reconnecting populations (including reduced inbreeding depression and increased adaptive capacity) with the perceived risk of outbreeding depression, which requires comprehensive knowledge of the landscape of adaptive, neutral, deleterious, and structural variation across a species' range. Using a combination of reduced-representation and whole-genome sequencing, we characterized genomic diversity and differentiation for the Arkansas Darter () across its range in the Midwestern US.
