Article Abstract

The goal of this study was to evaluate P450 aromatase localization in the epididymis of two different vertebrates: the lizard Podarcis sicula, a seasonal breeder, and Rattus rattus, a continuous breeder. P450 aromatase is a key enzyme involved in the local control of spermatogenesis and steroidogenesis and we proved for the first time that this enzyme is represented in the epididymis of both P. sicula and R. rattus. In details, P450 aromatase was well represented in epithelial and myoid cells and in the connective tissue of P. sicula epididymis during the reproductive period; instead, during autumnal resumption this enzyme was absent in the connective tissue. During the non-reproductive period, P450 aromatase was localized only in myoid cells of P. sicula epididymis, whereas in R. rattus it was localized both in myoid cells and connective tissue. Our findings, the first on the epididymis aromatase localization in the vertebrates, suggest a possible role of P450 aromatase in the control of male genital tract function, particularly in sperm maturation.

Human cytochrome P450 enzyme CYP19A1, commonly referred to as aromatase, plays a critical role in estrogen biosynthesis by catalyzing the conversion of C19 androgens into aromatic C18 estrogens. Alterations in aromatase activity have been implicated in the development and progression of estrogen-dependent diseases. Genetic mutations, including nonsynonymous single nucleotide polymorphisms (SNPs), can markedly impact the catalytic function of CYP19A1.

Gonadal development in Ruditapes philippinarum involves germ cell formation, sex differentiation, vitellogenesis, and maturation, regulated by complex genes-hormone interactions. To elucidate these mechanisms, multi-omics analyses (transcriptomics, proteomics, and steroid targeted metabolomics) and qPCR were applied to gonads across developmental stages in clams. Differential expression analysis revealed 332 testicular and 560 ovarian genes, alongside 174 testicular and 107 ovarian proteins.

Polycyclic aromatic hydrocarbons (PAHs), considered endocrine disruptors, affect the animals reproduction by interfering with the production of sex steroids. In the present study, the in vitro effects of selected PAHs (naphthalene, phenanthrene and BaP) on steroid production of cultivated ovarian cells from Arabian sea bream (Acanthopagrus arabicus) was assessed. Ovarian cells were cultured in Leibovitz L-15 (L-15) medium containing naphthalene, phenanthrene and benzo(a)pyrene with or without androstenedione (AD) as precursors and17b-estradiol (E2), progesterone (P4) and testosterone (T) were then measured.
