GABA and 5-HT agonists are effective in attenuating the behavioral effects of psychostimulants. However, they induce adverse side effects when used in high doses. The previous evidence has suggested that the 5HT receptor activation effect could be produced by an increased release of GABA in the ventral tegmental area (VTA) and the consequent activation of GABAergic receptors. Therefore, the objective of this study was to evaluate the effects of joint administration of an intermediate dose of the GABA agonist baclofen (3.0 mg/kg) with different doses of the 5HT agonist Ro60-0175 (0.3, 1.0, and 3.0 mg/kg) on the locomotor sensitization expression induced by the repeated administration of amphetamine (1.0 mg/kg). Our results showed an attenuation of the expression of sensitization in a dose-dependent manner with both agonists. In both cases, we observed a complete blockade at the highest dose. In addition, the intermediate dose of baclofen increased the effects of the three doses of Ro60-0175. These results support the role of the joint action of GABA and 5-HT receptors in the effects of psychostimulants. However, it remains to be explored whether the observed effect can be attributed to receptors located in the VTA or the nucleus accumbens.

Adolescent binge drinking is a risk behavior associated with the development of neuropsychiatric disorders later in life, but the pathophysiological mechanisms rendering the adolescent brain vulnerable to the long-term consequences of heavy alcohol consumption are only partially understood. Here, we used a mouse model of adolescent binge drinking and focussed on G protein-gated inwardly rectifying potassium (GIRK) channels which are a molecular target of both ethanol and the pluripotent growth and differentiation factor activin A. In whole-cell recordings from dentate gyrus granule cells in brain slices from alcohol-naive mice, we found a striking reversal of the effect of activin A on ethanol-evoked GIRK current as the mice matured: Whereas activin A reduced the ethanol response in cells from adult mice, the already lower ethanol threshold in cells from young mice was brought down even further by activin A.

Importance: Aripiprazole, a partial D2 receptor agonist, is proposed to enhance prefrontal cortex (PFC) dopamine function, improving working memory and GABA transmission, which supports social functioning. Long-acting injectable (LAI) antipsychotics are known to improve patient adherence, leading to enhanced long-term effects on behavioral outcomes.
