Article Abstract

To overcome the limitations of fast scan cyclic voltammetry (FSCV) to discriminate between catecholamines, we discuss new approaches to monitor the dynamics of these neurochemicals with high spatial, genetic, and temporal specificity.

Background: Takotsubo cardiomyopathy (TC) frequently co-occurs with spontaneous coronary artery dissection, but its co-occurrence with extracoronary arterial dissections is rare.

Case Summary: A 68-year-old woman with hypertension, hyperlipidemia, and type 2 diabetes reported lower abdominal pain and vomiting amid severe emotional distress. Within hours of admission to the hospital, she developed acute coronary syndrome and was diagnosed with TC, which was managed with dual antiplatelet therapy.

Adaptive responses to challenging environments depend on optimal function of the locus coeruleus (LC), the brain's main source of noradrenaline and primary mediator of the initial stress response. Combining functional circuit dissection and causal in vivo interventions in mice, we here investigate a built-in peptidergic regulatory system that restricts LC noradrenergic output. In particular, we characterize a population of neuropeptide Y (NPY)-expressing neurons surrounding LC noradrenergic cells.

Background: Calcitonin gene-related peptide (CGRP) release from trigeminal structures is central to migraine pathophysiology. This study employed an ex vivo model preserving anatomical continuity between the trigeminal nucleus caudalis (TNC) and trigeminal ganglion (TG) to investigate (1) whether TNC stimulation induces peripheral CGRP release from the TG and (2) the potential involvement of a distinct dopamine-endocannabinoid mechanism.

Methods: Tissues were dissected as a single unit and placed in custom 3D-printed chambers, allowing targeted stimulation of either the TNC or the TG while measuring CGRP in both compartments.

Emerging evidence highlights the gut microbiota as a pivotal determinant of pharmacological efficacy. While ()-derived tyrosine decarboxylases () are known to decarboxylate levodopa (L-dopa), compromising systemic bioavailability, the causal mechanisms underlying microbiota-mediated pharmacodynamic variability remain unresolved. In our study, we employed antibiotic-induced microbiota depletion and fecal microbiota transplantation (FMT) to interrogate microbiota-L-dopa interactions in MPTP-induced Parkinson's disease (PD) mice.
