Phospholipase C (PLC) enzymes produce second messengers that increase the intracellular Ca concentration and activate protein kinase C (PKC). These enzymes also share a highly conserved arrangement of core domains. However, the contributions of the individual domains to regulation are poorly understood, particularly in isoforms lacking high-resolution information, such as PLCϵ. Here, we used small-angle X-ray scattering (SAXS), EM, and functional assays to gain insights into the molecular architecture of PLCϵ, revealing that its PH domain is conformationally dynamic and essential for activity. We further demonstrate that the PH domain of PLCβ exhibits similar dynamics in solution that are substantially different from its conformation observed in multiple previously reported crystal structures. We propose that this conformational heterogeneity contributes to subfamily-specific differences in activity and regulation by extracellular signals.

Warfarin is a widely used vitamin K antagonist (VKA) with known pleiotropic effects beyond anticoagulation. Preclinical and case-control evidence suggests that warfarin may affect hematopoiesis, but longitudinal human evidence is lacking. To explore this potential effect, we conducted a post-hoc analysis of participants in the Hokusai-VTE and ENGAGE AF-TIMI 48 trials, which randomized patients to warfarin or the direct oral anticoagulant edoxaban with routine laboratory testing at predefined follow-up visits.

This study investigates the HO and CO sorption behavior of two chemically distinct polystyrene-divinylbenzene-based ion exchange sorbents: a primary amine and a permanently charged strong base quaternary ammonium (QA) group with (bi)carbonate counter anions. We compare their distinct interactions with HO and CO through simultaneous thermal gravimetric, calorimetric, gas analysis, and molecular modeling approaches to evaluate their performance for dilute CO separations like direct air capture. Thermal and hybrid (heat + low-temperature hydration) desorption experiments demonstrate that the QA-based sorbent binds both water and CO more strongly than the amine counterparts but undergoes degradation at moderate temperatures, limiting its compatibility with thermal swing regeneration.

Purpose: To reassess the anatomic basis of optic disc pit maculopathy (OPM) using swept-source optical coherence tomography (SS-OCT) and to characterize the broader structural abnormalities comprising the optic pit complex.
Methods: Sixteen patients with OPM were imaged using a high-resolution SS-OCT system (DREAM OCT). Cross-sectional and volume-rendered scans were analyzed for lamina cribrosa defects, intraneural cavitations, and pathways for fluid entry into or beneath the retina.

Background: Appendiceal adenocarcinomas and low-grade appendiceal mucinous neoplasms (LAMNs) are rare tumours. Much of the existing knowledge is derived from registry-based studies, particularly the Surveillance, Epidemiology, and End Results database in the USA.
Methods: This retrospective cohort study used data from the Swedish Cancer Registry, Swedish Cause of Death Registry, and the National Patient Registry to analyse demographic characteristics and outcomes of patients diagnosed with appendiceal adenocarcinoma or LAMN between 2005 and 2019.
