Article Abstract

The socioeconomic burden of chronic back pain related to intervertebral disc (IVD) disease is high and current treatments are only symptomatic. Minimally invasive strategies that promote biological IVD repair should address this unmet need. Notochordal cells (NCs) are replaced by chondrocyte-like cells (CLCs) during IVD maturation and degeneration. The regenerative potential of NC-secreted substances on CLCs and mesenchymal stromal cells (MSCs) has already been demonstrated. However, identification of these substances remains elusive. Innovatively, this study exploits the regenerative NC potential by using healthy porcine NC-derived matrix (NCM) and employs the dog as a clinically relevant translational model. NCM increased the glycosaminoglycan and DNA content of human and canine CLC aggregates and facilitated chondrogenic differentiation of canine MSCs . Based on these results, NCM, MSCs and NCM+MSCs were injected in mildly (spontaneously) and moderately (induced) degenerated canine IVDs and, after six months of treatment, were analyzed. NCM injected in moderately (induced) degenerated canine IVDs exerted beneficial effects at the macroscopic and MRI level, induced collagen type II-rich extracellular matrix production, improved the disc height, and ameliorated local inflammation. MSCs exerted no (additive) effects. In conclusion, NCM induced regenerative effects on degenerated canine IVDs. NCM may, comparable to demineralized bone matrix in bone regeneration, serve as 'instructive matrix', by locally releasing growth factors and facilitating tissue repair. Therefore, intradiscal NCM injection could be a promising regenerative treatment for IVD disease, circumventing the cumbersome identification of bioactive NC-secreted substances.

Introduction: Polylaminin, an improved form of the natural protein laminin, has been shown to promote axonal regeneration and functional recovery in animal models of acute spinal cord injury (SCI), and is safe and potentially beneficial in humans when administered within the first days after traumatic SCI. This study aimed to evaluate the effect of polylaminin in dogs with chronic SCI.

Methods: We conducted a prospective, longitudinal study in six paraplegic dogs with severe chronic thoracolumbar SCI (T3-L3) caused by trauma ( = 2) or disc degeneration ( = 4).

Recently, RNA has shown great potential for use in forensic genetics. Our previous work indicated that hair shafts possess detectable RNA levels. Fallen hair samples at crime scenes are common, but human identification is difficult because of the degeneration of traceable nuclear DNA.

Although rare, tick-borne encephalitis (TBE) is one of the most important and commonly fatal viral diseases affecting the central nervous system (CNS). This arboviral disease is transmitted by ticks and prevalent in widespread parts of Eurasia. Besides humans, several domestic animals such as dogs, horses, and ruminants can also be infected.

Background: Client-owned dogs presenting clinically with intervertebral disc disease (IVDD) are a potential comparative animal model to help advance the understanding of disc degeneration and its treatment. To utilize dog patients as a model, noninvasive imaging techniques are needed that can characterize subtle and progressive changes in disc health in longitudinal and treatment efficacy studies. The purpose of this study was to assess the sensitivity of quantitative MRI techniques in detecting disc degeneration in client-owned, nonchondrodystrophic-breed dogs.

Trypanosoma cruzi DTU TcI disproportionately associated with fatal T. cruzi myocarditis in dogs in Texas, USA.

Acute heart failure and sudden death are possible outcomes of Trypanosoma cruzi infection in domestic dogs. Understanding which discrete typing units (DTUs) are present in fatal canine Chagas disease cases may help explain variation in disease outcome among infected dogs. We conducted a retrospective study of cases from the Texas A&M Veterinary Medical Diagnostic Laboratory and sampled heart tissue from 62 dogs with a histopathologic diagnosis of Chagas disease from 2007 to 2013 for which formalin-fixed paraffin embedded heart tissue was available.
