Sperm entry in mammalian oocytes triggers intracellular Ca oscillations that initiate resumption of the meiotic cell cycle and subsequent activations. Here, we show that phospholipase C zeta 1 (PLCζ1) is the long-sought sperm-borne oocyte activation factor (SOAF). Plcz1 gene knockout (KO) mouse spermatozoa fail to induce Ca changes in intracytoplasmic sperm injection (ICSI). In contrast to ICSI, Plcz1 KO spermatozoa induced atypical patterns of Ca changes in normal fertilizations, and most of the fertilized oocytes ceased development at the 1-2-cell stage because of oocyte activation failure or polyspermy. We further discovered that both zona pellucida block to polyspermy (ZPBP) and plasma membrane block to polyspermy (PMBP) were delayed in oocytes fertilized with Plcz1 KO spermatozoa. With the observation that polyspermy is rare in astacin-like metalloendopeptidase (Astl) KO female oocytes that lack ZPBP, we conclude that PMPB plays more critical role than ZPBP in vivo. Finally, we obtained healthy pups from male mice carrying human infertile PLCZ1 mutation by single sperm ICSI supplemented with Plcz1 mRNA injection. These results suggest that mammalian spermatozoa have a primitive oocyte activation mechanism and that PLCζ1 is a SOAF that ensures oocyte activation steps for monospermic fertilization in mammals.

The present study aimed to (1) evaluate the effects of different concentrations of the polysaccharide extract of Cissus sicyoides (PE-Cs) during in vitro culture of preantral follicles included in goat ovarian tissue on (i) follicular morphology and activation, (ii) ovarian stromal density, (iii) follicular and oocyte diameters, (iv) antioxidant enzymes activity (SOD, CAT, and GPx), (v) quantification of MDA, thiol, and nitrite levels; as well as to (2) measure the total antioxidant capacity of the extract. The ovarian cortex fragments were cultured at 39 °C in a humidified atmosphere with 5% CO for 6 d in alpha-modified minimum essential medium (αMEM) supplemented with insulin, transferrin, and selenium; hypoxanthine; glutamine; and bovine serum albumin, which was called αMEM alone or added of PE-Cs at 20, 40, or 80 µg/mL. At the end of the culture period, a reduction in the percentage of normal follicles in all treatments using PE-Cs compared to fresh control and αMEM.

This case report describes the use of doxazosin (Cardura) as a treatment for a patient with an autosomal dominant , single-nucleotide R398Q pathogenic variant, which has not previously been described in the literature. The patient has gain-of-function pathogenic genetic variant. Because of the patient's continued seizure burden with the use of traditional antiseizure medications and failed invasive antiseizure interventions, an oocyte cell line with the specific genetic variant was created to test efficacy of various medications.

Background: Lysosomes serve not only in the degradation of cellular components but also as calcium (Ca) stores. In this study, we investigated the effects of trans-Ned19, an inhibitor of lysosomal calcium channels known to block two-pore channels (TPCs), on fertilization and oocyte activation in mice.
Methods: Pronuclear formation was assessed via Hoechst 33342 staining, cortical granule release was evaluated using agglutinin-fluorescein isothiocyanate (LCA-FITC) staining, intracellular Ca levels were monitored with Cal-520 AM, and sperm motility was analyzed using a sperm motility analysis system (SMAS).

Immature zebrafish oocytes are highly susceptible to high temperatures, making it difficult to warm cryopreserved oocytes rapidly. In the present study, we aimed to investigate whether thermosensitive channels, lipid mediators, and ferroptosis are involved in heat stress-induced injury in immature zebrafish oocytes. Oocytes were injected with inhibitors of a heat-sensitive channel (TRPV1) and multiple enzymes-cytosolic phospholipase Aα (cPLAα), cyclooxygenases (COXs), arachidonate lipoxygenase 5 (ALOX5), and lysophosphatidylcholine acyltransferase 2 (LPCAT2).
