Altered glucose reabsorption the facilitative glucose transporter 2 (GLUT2) during diabetes may lead to renal proximal tubule cell (RPTC) injury, inflammation, and interstitial fibrosis. These pathologies are also triggered by activating the cannabinoid-1 receptor (CBR), which contributes to the development of diabetic nephropathy (DN). However, the link between CBR and GLUT2 remains to be determined. Here, we show that chronic peripheral CBR blockade or genetically inactivating CBRs in the RPTCs ameliorated diabetes-induced renal structural and functional changes, kidney inflammation, and tubulointerstitial fibrosis in mice. Inhibition of CBR also downregulated GLUT2 expression, affected the dynamic translocation of GLUT2 to the brush border membrane of RPTCs, and reduced glucose reabsorption. Thus, targeting peripheral CBR or inhibiting GLUT2 dynamics in RPTCs has the potential to treat and ameliorate DN. These findings may support the rationale for the clinical testing of peripherally restricted CBR antagonists or the development of novel renal-specific GLUT2 inhibitors against DN.

The endocannabinoid system (ECS) plays a critical role in fear learning and maintenance and may, therefore, be implicated in the pathophysiology of posttraumatic stress disorder (PTSD). The exact role of cannabinoid receptor 1 (CB1R), a key component of the ECS, remains unclear. Although preclinical studies largely suggest CB1R downregulation in PTSD, the only prior study of CB1R availability in individuals with PTSD reported higher levels than in controls.

Background: Thoracic aortic dissection (TAD) is a fatal cardiovascular disease characterized by vascular smooth muscle cell (VSMC) phenotypic remodeling and inflammation. Cannabinoid 1 (CB1/Cnr1) receptor has been implicated in cardiovascular diseases, but its role in TAD and its downstream signaling remains unclear. Genistein (Gen), an isoflavone abundant in legumes, has demonstrated promising therapeutic potential in cardiovascular diseases such as vascular calcification, hypertension, and atherosclerosis due to its anti-inflammatory, antioxidant, and metabolic regulatory properties.

Blockade of endocannabinoid CB1R has shown effects on adulthood sociability, memory and fear responses, under basal and stress conditions. In this study, we examined sex-specific effects of exposure to heterotypic stress in the prepubescent period, marked by maturation of brain systems, on endocannabinoid receptors expression, corticosterone secretion and effects of CB1 receptor blockade on social and cognitive responses. Sixty-four (N = 32 per sex) adolescent male and female rats were randomly assigned to a stress or no stress condition.

A series of new N,N'-diarylureas is reported as potential cannabinoid-1 (CB-1) receptor inhibitors. The synthesis of the new N,N'-diarylureas is achieved from the reaction of two substituted anilines with the aid of triphosgene. One aniline carries a pyrazol-1-yl or 1H-1,2,3-triazolyl or 2H-1,2,3-triazolyl propan-2-one group at position-3, while the other aniline is substituted by fluoro, bromo, methoxy, cyano, morpholino, or 4-methyl-2-nitro groups.
